Objective:Methods Animal model was established by injectingβ-amyloid protein into paracele of adult rats under stereotaxic apparatus,Lead to Morris water maze test learning and memory impairment in rats. To APP, BACE and Aβ1-40 reflect the"Three olfactory acupoints"on the ADβ-amyloid formation during the intervention effect.Methods:60 adult male SD rats were randomly divided into six groups(each 10 rats):Ⅰ"Three olfactory acupoints"+ AD+olfactory nerve cut model group;Ⅱ"Three olfactory acupoints"+ AD model group;Ⅲolfactory nerve cut + AD model group;ⅣAD model group;Ⅴolfactory nerve cut group;Ⅵblank control group.Ⅰ,Ⅱ,ⅢandⅣgroups were established by injectingβ-amyloid protein into paracele under stereotaxic apparatus;ⅤandⅥgroups were established by injecting strokephysiological saline solution into paracele under stereotaxic apparatus.ⅠandⅡgroups were given " Three olfactory acupoints "treatment the second day, one time per day, fifteen days a course ,continuing five courses, rest tow days per course. The other groups were conventional breeding to the end of treatment. In the end of treatment, animals trained for Morris water maze for one week. All animals were sacrificed at the end of Morris water maze. Immunofluorescence staining observes the Aβ1-40 positive cell rate in hippocampus of rats. Western-blot observes, APP and BACE protein content in hippocampus of rats.All data entered into a computer, using SPSS16.0 statistical package for processing.Results:The Morris water maze test: Compared with AD model group, the escape latency decreased significantly (P<0.05) in the groups treated with"Three olfactory acupoints". Compared with"Three olfactory acupoints"+ AD model group, the escape latency of"Three olfactory acupoints"+ AD+ olfactory nerve cut model group increased significantly(P<0.05). Immunohistochemistry test: Compared with model group, the number of positive cells decreesed significantly (P<0.05) in the groups treated with"Three olfactory acupoints". Compared with"Three olfactory acupoints"+ AD model group, the number of positive cells of"Three olfactory acupoints"+ AD+ olfactory nerve cut model group increased significantly(P<0.0 5). The western blot expression of APP and BACE were according with immune- histochemistry.Conclusion:The results of this study show that the"Three olfactory acupoints"treatment of AD through the mechanism of the olfactory pathway has enriched the idea of acupuncture treatment of AD.
