Objective:To investigate the effects of cisplatin and 5-Fu on the proliferation and glycolytic signature of MKN-45 and SGC-7901 gastric cancer cells. Methods: Proliferation of MKN-45 and SGC-7901 cells before and after chemotherapy drugs (5-Fu or cisplatin) were measured by MTT assay. Cells were harvested, counted, and seeded into 96-well microtiter plates (5×103 cells/well) and incubated overnight. And then they were treated with either vehicle or various concentrations of 5-Fu or DDP. The morphology of cells was observed in inverted phase contrast microscopy. The changes of the key glycolytic elements (pyruvate kinase PKLR, PKM2, a-enolase, Aldolase A and Heat shock protein 27) in MKN-45 and SGC-7901 cell lines under treatment of 5-Fu and DDP were analyzed by Western blotting. Results:(1) The SGC-7901 cell line was insensitive to 5-Fu and cisplatin, and had no obvious inhibition. (2) The MKN-45 cell line was sensitive to cisplatin while insensitive to 5-Fu, and in a concentration-dependent manner at certain range of cisplatin concentrations. (3) Following cisplatin treatment, the levels of some key glycolytic elements (pyruvate kinase PKLR, PKM2,α-enolase, Aldolase A and Heat shock protein 27) in the MKN-45 cells were decreased. (4) On the contrary, when the MKN-45 cells were exposed to 5-Fu, the expression of those proteins increased slightly. (5) After exposed to 5-Fu or cisplatin, the expressions of the key glycolytic elements in the SGC-7901 cell line were slightly raised with the increasing doses of 5-Fu or cisplatin. Conclusion:gastric carcinoma cell lines, SGC-7901 and MKN-45, have different sensitivity to 5-Fu and cisplatin, and SGC-7901 have a certain resistance to chemotherapy while the MKN-45 cell line was sensitive to cisplatin and in a concentration-dependent manner at certain range of cisplatin concentrations. so we should be careful in clinical application.
