Purpose:Through the experimental study on effect of Astragalus combined with cisplatin on gastric cancer cell line SGC-7901, clarify the positive effect of Astragalus play against cancer, further from the anti-tumor effect and mechanism of cell and molecular biology on Chinese Medicine Radix astragaliMaterial and method:1. The object of study:human gastric cancer cell line SGC-7901, the logarithmic growth phase of the cells used in the experiments.2. Grouping:blank group (cells with culture medium without adding cisplatin), control group (single cisplatin2ug/ml), Astragalus1group (cisplatin2ug/ml plus Astragalus lg/ml).Astragalus2group (cisplatin2ug/ml plus Astragalus0.5g/ml) Astragalus3group (cisplatin2ug/ml plus Astragalus0.25g/ml), Astragalus4group (cisplatin2ug/ml plus Astragalus0.125g/ml).3. Test method:3.1The effect of different groups on proliferation of human gastric cancer cell line SGC-7901by MTT method, enzyme mark instrument request each hole measured at the wavelength of490nm degree (A), according to the A to estimate the effect of drugs on cell proliferation.3.2Expression of Bcl-2and ICAM-1gastric cancer cell line SGC-7901by RT-PCR, the last half quantitative analysis.Results:1. MTT method to measure the impact of different groups of SGC-7901cell line proliferation experimental results:Compared with the control group, the cisplatin group and so on dose cisplatin with different concentrations of Astragalus combined group can inhibit gastric cancer SGC7901cell line value-added and time-dependent manner,36h after cisplatin group inhibit cell proliferation slowed. Gradually increased with the concentration of astragalus, the inhibition was significantly enhanced. Compared with the cisplatin group, the high concentration of Astragalus cisplatin mixture (the astragalus1g/ml,0.5g/ml)24h SGC7901gastric cancer cell lines significantly inhibited than to be significantly stronger than cisplatin concentration Astragalus cisplatin mixture (Astragalus0.25g/ml)24h SGC7901gastric cancer cell lines significantly inhibited in the cisplatin group, low concentrations of Astragalus of cisplatin mixture (Astragalus0.125g/ml)24h significantly inhibited gastric48h stronger, but with Shun platinum-group differences.2. RT-PCR method to measure gastric cancer SGC-7901cell lines the Bcl-2and the expression of ICAM-1The expression of ICAM1in Astraglus membranaceus and cisplatin combination group is higher than singgle SGC7901gastric cancer cell lines. The expression of ICAM1in high-dose cisplatin astaglatin groups mixture group (astragalus membranaceus1g/ml,0.5g/ml) is higher than cisplatin group. The expression of BCL-2in Astraglus membranaceus and cisplatin combination group is lower than singgle SGC7901gastric cancer cell lines. The expression of bcl-2in high-dose cisplatin astaglatin groups mixture group (astragalus membranaceus1g/ml,0.5g/ml) is lower than cisplatin group.Conclusion:1. High concentrations of the Astragalus cisplatin group (Astragalus lg/ml,0.5g/ml) for different time points the gastric SCG7901cells significantly inhibited strains.2. Astragalus membranaceus plus cisplatin group and cisplatin group can significantly increase the expression of ICAM1in human gastric cancer cell line SGC-7901;compare with cisplatin group, increased expression of ICAM1of the effect of Astragalus membranaceus plus cisplatin group in human gastric cancer cell line SGC-7901was superior. 3. Astragalus membranaceus plus cisplatin group and cisplatin group can significantly reduce the expression of BCL-2in human gastric cancer cell line SGC-7901; compare with cisplatin group, reduce expression of BCL-2of the effect of Astragalus membranaceus plus cisplatin group in human gastric cancer cell line SGC-7901was...
