To observe the pathological changes and influence on the level of chlamydial heat shock protein 60 (CHSP60),tumor necrosis factor-alpha(TNF-a) and interferon-γ(IFN-λ) on fallopian tube epithelium by intravaginal inoculation of Salvia miltiorrhiza injection. To observe mechanism of action of Salvia miltiorrhiza injection on salpingitis infertility patients after interventional recanalization.MethodsForty four female SD rats, ranging from 8-10 weeks of age, were divided into Blank group four and experimental group Forty. experimental group were inoculated progesterone by intramuscular injection daily, and were inoculated 50ul×106 of inclusion-forming units of Chlamydia trachomatis E serovar on right uterine horn after one week and then were inoculated progesterone by intramuscular injection every other day, inoculated progesterone 7 times totally. Aftet infection with Ct, forty SD rats were randomly divided into medium does of Salvia miltiorrhiza(Sm) group, high does of Salvia miltiorrhiza (Sm) group, Western medicine (Wm) group and Normal saline (Ns) group according intravaginal inoculation of different drugs and different drugs. Four groups of rats were inoculated certain drugs and drugs intravaginally once every three days.After intravaginlly inoculated 5 times, the histopathologic changes of salpingitis in rats were observed and immunohistochemical expression of CHSP60,TNF-αand IFN-λwere observed. The clinical cases, which were diagnosed as salpingitis infertility and accepted interventional recanalization successfully, were randomly divided into SM group and Wm group. The treatment effect of two groups were compared. We tride to explain the mechanism of Chinese medicines for prmoting blood circulation,such as Salvia miltiorrhiza, through animal experiments and clinical research.ResultThe unaided eye:Compared the medium does of Sm group and high does of Sm group, Connective tissue aroud the uterus and the pelvic wall adhesions were reltively serious in Wm group and Ns group. In Ns group, bilateral uterus of three rats becomed thickening and pale. In Wm group, bilateral uterus of only one rats becomed thickening and pale. However, in medium does of Sm group and high does of Sm group, none of uterus becomed thickening and pale.Lighet microscope:Compared the number of neutrophils in bilateral uterine wall of the experimental group at high magnification:there was no significant difference between the Wm group and the Ns group and also between the medium does of Sm group and high does of Sm group, P>0.05. However, the number of neutrophils in the medium does of Sm group and high does of Sm group were statistically lower, compared with Ns group and Wm group, P<0.05. Compared the number of inclusion-forming units in right uterine wall of the experimental group at high magnification:there was no significant difference between the Wm group and the Ns group and also between the medium does of Sm group and high does of Sm group, P>0.05. However, the number of inclusion-forming units in the medium does of Sm group and high does of Sm group were statistically lower, compared with Ns group and Wm group, P<0.05. Compared the level of immunohistochemical expression of CHSP60,TNF-αand IFN-λ:The experimental group were statistically higher than blank group, P<0.05. there was no significant difference between the Wm group and the Ns group and also between the medium does of Sm group and high does of Sm group, P>0.05. However, the level of immunohistochemical expression of CHSP60,TNF-αand IFN-λin the medium does of Sm group and high does of Sm group were statistically lower, compared with Ns group and Wm group, P<0.05.Clinic research:Compared with Wester medicine, Chinese medicines for prmoting blood circulation, such as Salvia miltiorrhiza, could help to improveintrauterine pregnancy rate after Interventional Recanalization ConclusionsInfection with Chlamydia trachomatis E serovar in SD rats can produce upper genital inflammation, such as hydrosalpinx and pelvic adhesions, and can make the level of CHSP60,TNF-αand IFN-λincreased significantly.Salvia miltiorrhiza injection, inoculated by intravaginally, reduce the secretion of inflammatory cells and suppresses the inflammation. We suppose that inhibition of CHSP60,TNF-αand IFN-λprobably suppresses the inflammationSm injection can keep the tubal patency and improve pregnancy rate for salpingitis infertility patients after interventional recanalization.
