Effect Of Salvia Miltiorrhiza Injection On Expression Of VEGF And Integrin αvβ3 In Endometrium

Objective:To study the expression of vascular endothelial growth factor(VEGF)and integrin αvβ3 in endometrium under the action of Salvia miltiorrhiza injection,and further explored the effect of Salvia miltiorrhiza injection on endometrial receptivity.Methods:Endometrial tissues were collected from 25 infertile patients undergoing laparoscopic combined exploration in Maternal and Child Health Hospital of Guiyang Province from January 2020 to December 2020,including 9 patients with fallopian tube obstruction,8 patients with PCOS,and 8 patients with endometriosis.Postoperative disease examination confirmed that the endometrium was at the hyperplasia stage in 13 cases,and the secretarial stage in 12 cases.The endometrium taken out under aseptic condition was isolated and cultured in vitro.The endometrial cells cultured to the third generation were cultured in different concentrations of Salvia miltiorrhiza injection(0mg/ml、0.5mg/ml、1mg/ml、2mg/ml、4mg/ml、8mg/ml、 16mg/ml)for 48 hours,and the cellular proteins were extracted for follow-up experiments.The expression of VEGF and integrin αvβ3 in different concentrations of Salvia miltiorrhiza injection was detected by Western-bloting.Results:VEGF and integrin αvβ3 were expressed in endometrium of all patients,and their expression levels were decreased in PCOS patients and EMS patients compared with those in patients with fallopian tube blockage(P<0.05).The expressions of VEGF and integrin αvβ3 in the secretory endometrium of each group were increased compared with those in thproliferative stage(P<0.05).The expression of VEGF and integrinαvβ3 showed an up-regulation trend after 48 hours of culture with Salvia miltiorrhiza injection,and the expression of VEGF and integrin αvβ3 showed a positive correlation,which was the highest when the drug concentration was 4mg/ml(P <0.05).Conclusions:1.The expression levels of VEGF and integrin αvβ3 in PCOS patients and EMS patients were lower than those in patients with fallopian tube blockage,suggesting that endometriosis and PCOS infertility patients have low endometrium receptivity.2.The expression of protein in secretory endometrium is higher than that in proliferative endometrium,suggesting that the expression of VEGF and integrin αvβ3is cycle specific.3.Under the action of Salvia miltiorrhiza injection,the expression of VEGF and integrin αvβ3 were up-regulated,it was most significant at 4mg/ml,suggestting that Salvia miltiorrhiza injection at a certain concentration may provide a new idea and method for improving endometrial receptivity.
