ObjectiveTo study the chemical composition in radix of Ilex asprella(Hook.et Arn.)Champ.ex Benth., and the quality of wild resource.It may help us to promote the pharmacological active substance research of Gangmei,provide a scientific basis of development of the quality standard and reasonable application,and ensure access to safe,effective,stable and reliable Gangmei.Methods1. Germplasm collection and identification of Gangmei.1.1 Investigation and collection of Gangmei We collecte Gangmei wild germplasm on the basis of surveys of wild resources.1.2 TLC reseach of GangmeiUse TLC to study roots.stems.leaves of Gangmei from different areas,Chloroform: Ethyl Acetate:Methanol:Water(15:40:22:10, place under 10℃,use the substrabe solution) and Cyclohexane:Acetic ether (5:1) as the developing solvent.1.3 IR reseach of GangmeiUse IR to study fingerprint of Gangmei.We compa-ed the roots and stems from different habitats and collection seasons of Gangmei by Direct analysis method, Similarity and Second derivative IR spectra.2. Chemical composition studyUse silicone,Sephadex LH-20,ODS and other chromatography to isolate chemical composition of Gangmei,and use spectral methods to identify the structures.3. Quality analysis and evaluation3.1 Check of GangmeiWe determined water content,heavy metal of Gangmei according to the first part of the Chinese pharmacopoeia 2010 appendix IX H,IX B,IX E. 3.2 ExtractWe determined extract content of Gangmei according to the first part of the Chinese pharmacopoeia 2010 appendix XA.3.3 content determinationWe determine total saponins content of Gangmei by UV-RIS,and determine pomolic acid content of Gangmei by HPLC.Result1. Germplasm collection and identification of Gangmei.1.1 Investigation and collection of GangmeiGangmei wild resources are mainly distributed in Guangdong, Guangxi and so on.We collected 14 wild germplasm,1 cultivated germplasm and 6 commercial drugs.1.2 TLC reseach of GangmeiThe results showed that the roots's chemical composition similar to the stems's,but the roots's and the stems's chemical composition are different from the leaves's.The roots of gangmei from different habitats are similarity.1.3 IR reseach of GangmeiWe compared the roots and stems of Gangmei by direct analysis method,and found that the two IR spectrum are different in 1737,1246 and 1508cm-1.We calculated similarity of Gangmei by computer assistant software.The roots and stems of Gangmei are different. The roots and stems of Gangmei from different collection seasons are different in Second derivative IR spectra.2. Chemical composition studyThere were 18 compounds isolated.Through Physical and chemical properties and spectral analysis identified 11 compounds'structure.6 compounds were first isolated from Gangmei.3. Quality analysis and evaluation3.1 Check of GangmeiThe results showed that water content<10%,As,Cu,Hg and total heavy metals within the allowable range of "import and export of medicinal plants and preparation of green industry standard", Pb and Cd exceeding the permitted range. 3.2 ExtractWater extract content of the roots of Gangmei should not be less than 4.0%,alcohol extract content should not be less than 2.7%. Water extract content of the stems of Gangmei should not be less than 3.0%, alcohol extract content should not be less than 2.0%.3.3 content determinationThe results showed that the total saponins content of roots of Gangmei were greater than the stems. The roots'total saponins were between 2.3%~7.4%, the stems'total saponins were between 0.9%~2.3%.Goods's total saponins content were more than the stems'.We determined pomolic acid content of Gangmei by HPLC. The results showed that the pomolic acid content of the roots of Gangmei were greater than the stems. The roots' pomolic acid content were between 0.23%~0.59%, the stems'pomolic acid content were between 0.011%~0.14%. Goods's pomolic acid content were more than the stems'.ConclusionThe present study reasearch the chemical composition of Gangmei,and use TLC,IR, water content,heavy metal, total saponins content and pomolic acid content to compare the diffence of the roots and stem of Gangmei, providing a scientific basis of development of the quality standard and reasonable application.
