Celosia argentea L. (Family-Amaranthaceae) is firstly recorded in Shen Nong'sHerbal. In folklore practice, the seeds of Celosia argentea L. has been used traditionallyfor removing liver-heat, improving eyesight, clearing wind-heat, lowering the bloodpressure and anti-inflammation, according to Chinese Pharmacopoeia (edition of 2010).In order to study the chemical constituents of Celosia argentea L. seeds and provideevidence for establishing the quality standards of Celosia argentea L., we isolated andpurified the chemical constituents of 30% and 60% fractions of Celosia argentea L. seedsby HSCCC-ELSD. The structures were identified on the basis of spectral analysis and bycomparison with the data reported in literature. As a result, 5 compounds were isolated andidentified by spectral analysis. Celosin C1, Celosin D1, Celosin E, Celosin F and Celosin Gare found to be new compounds.This study also investigated the antitumor activities of Celosin A, Celosin B, CelosinC1, Celosin D1, Celosin E, Celosin F, Celosin G and Cristatain. The five tumor cell linesare: SHG44,HCT116,CEM,MDA-MB-435 and HepG2.IC50 of Celosin A, Celosin B andCristatain: SHG44(35.37±1.36×10-6 mol/L,21.65±1.16×10-6 mol/L,35.82±2.96×10-6mol/L),HCT116(38.69±1.86×10-6 mol/L,36.31±3.69×10-6 mol/L,40.36±4.11×10-6mol/L),CEM(39.82±2.74×10-6 mol/L,24.43±1.46×10-6 mol/L,47.76±2.66×10-6mol/L),MDA-MB-435(38.72±3.24×10-6 mol/L,12.68±1.08×10-6 mol/L,41.74±2.93×10-6mol/L),HepG2(39.53±2.94×10-6 mol/L,31.86±3.55×10-6 mol/L,42.82±2.32×10-6 mol/L).The result indicated that Celosin A, Celosin B and Cristatain have certain antitumoractivities. Celosin C1, Celosin D1, Celosin E, Celosin F and Celosin G have no antitumoractivities.The present study investigated the hepatoprotective effects of Celosin C1 and CelosinD1 against the CCl4-induced liver injury. The mice were administrated orally with totalethanol extract at 2.0mg·kg-1-8.0mg·kg-1 doses, respectively, for 7 days. On 7th day, 0.1%CCl4 were given to the mice 1 hour after the last administration. At 18 hours after CCl4injection, blood samples were drawn from the retro-orbital plexus. The results showed thatCelosin C1 and Celosin D1 remarkably lowered the serum AST and ALT compared to theCCl4-treated group (P<0.01).This study lays a foundation for the utilization of Celosia argentea L.
