Objective: To establish the animal model of stress urinary incontinence ( SUI)through imitating the birth trauma and the decrease of estrogen level combined with cutting off the pudendal nerves,then by identification and compared between models to offer reliable experiment object for research of SUI.Methods: Forty healthy female Wistar rats were randomly divided into four groups separated to 4 groups: GroupⅠwas the control group, GroupⅡwas the group of colpectasis and ovariectomy , GroupⅢwas the group of bilateral PNT, GroupⅣwas GroupⅡ+ GroupⅢ,each group contained 10 rats. the urodynamics examinations of these 40 rats were carried out post-operation on Week 1,Week 4,Week 8. The sneeze test done to confirm the presence of SUI on week 1. And histological examinations were carried out on week 8.Results:①The positively rats in sneeze test in groupⅡ,Ⅲ,Ⅳwere 6,9,10,respectively , but none of rats was positive in groupⅠ.②The leakage of urine point pressure in groupⅢandⅣwere significantly lower than those in groupⅠandⅡ(P<0.01),And in groupⅡwas lower than in groupⅠ(P<0.05); However,there was no significant difference between groupⅢand groupⅣ(P>0.05).③Histological examinations showed all layers of Urethra getting thinner, the structure is not obvious and atrophy of muscle layers as well as decreased innervation in groupⅢandⅣthan in groupⅠandⅡ.the content ofα-actin in the muscle of pelvic floor in groupⅣwere significantly lower than that in groupⅠ,ⅡandⅢ(P<0.01),And in groupⅢwas lower than in groupⅠ,Ⅱ(P<0.05),there was no significant difference between groupⅠand groupⅡ(P>0.05).Conclusion : Integrated colpectasis combined with bilateral oophorectomy and bilateral PNT can successfully set up SUI animal model and the model shows higher successful rate and satisfactory stability, simulates the occurrence of SUI in multiple factors exhaustively, has a characteristic of closer to the mechanism for human SUI.
