[Objective]By the vitro culture of human periodontal ligament fibroblasts (PDLFs) and helicobacter pylori (H.pylori),establishing the model of H.pylori infects PDLFs in vitro,observe the cell proliferation and changes of cell cycle after H.pylori effects on PDLFs,to probe into the relationship between H.pylori and PDLFs, and to provide laboratory evidence for the correlation between H.pylori and periodontal disease.[Methods]1.Vitro culture PDLFs,to adjust the cell concentration to 2×105/ml.2.Make use of selective culture of brandt modified egg yolk agar,culture H.pylori,and adjust the concentration of H.pylori to 1×108cfu/ml,2×107cfu/ml,4×106cfu/ml,8×105cfu/ml,0cfu/ml.3.Coculture different concentrations of H.pylori and PDLFs respectively,build the model of in vitro H.pylori infects PDLFs,adopt (Methyl thiazolyl tetraxolium,MTT) and (Flow cytometry,FCM),observe the effect of different concentrations of H.pylori on the cell proliferation inhibition and the cell cycle influence of PDLFs.[Results]After different concentrations of H.pylori affected PDLFs for 6hs,12hs,24hs,compared with the control group,OD value all reduced to (p<0.05),which illustrates that different concentrations of H.pylori can inhibit the proliferation of PDLFs. With the increase in the concentration of H.pylori,OD value reduces gradually;With the same concentration, as time goes on, OD value also reduces. After H.pylori affected PDLFs for 12h,with the increase in the concentration of H.pylori,the proportion of GoG1 in cell cycle gradually increased, the proportion of S and G2M gradually decreased to (P<0.05).[Conclusions]H.pylori has proliferation and cell cycle inhibition effect on PDLFs,H.pylori and periodontal disease are highly interrelated.
