Objective:Detecting the expression of the rat cochlear spiral ganglion neurons (SGN) NMDA receptor subunit gene and explore the effect of salicylate on the expression of the subnit NR1 of NMDA receptor in rat spiral ganglion neurons,To find the role of sodium salicylate ototoxicity mechanisms.Methods :Detected the expression of the of NMDA receptor subunits of the cultured tissues of the rat spiral ganglion neurons using Real-time quantitative reverse transcription-Polymerase chain reaction(FQ-PCR),then Detected the expression of the subunit NR1 of NMDA receptor after treatment with medium containing 5mM/L sodium salicylate for 2h,4h,6h and8h.Results:The normal rat SGN NMDA receptor subunit NR2B has the highest expression followed by NR1,NR3A,NR2D,NR3B,NR2A,NR2C.Except NR2D and NR3B the difference between two subunits has statistically significant(p<0.01); Compare to the control group,treatment group spiral ganglion neurons NR1mRNA NMDA receptor subunit expression was significantly higher (P <0.01) at each time point and the expression was positive correlations with the treatment time.Conclusion: The NMDA receptor subnits mRNA were expressed in the rat spiral ganglion neurons and the expression level was different; Sodium salicylate make increased the expression of the subnit NR1 mRNA of NMDA receptor in rat spiral ganglion neurons.
