Objective:Take Rhizoma Cyperi as an example and the preparation of the molecular weight< 1000da extract for the end product to probe into the basic mode on extraction of available substances in traditional Chinese medicine that contain volatile oils by semi-bionic extraction (SBE).Methods:①First, Rhizoma Cyperi decoction pieces were researched on source, character identification and quality standard;②Rhizoma Cyperi in the optimized conditions of supercritical fluid extraction based on the use of the same pieces, equipment, technical parameters for SFE-CO2 extraction, save the other extract, the residue uniform design method to extract molecular weight< 1000Da, HPLC total area of indicators, comprehensive evaluation, optimization of the preparation of molecular weight< 1000Da SBE extract conditions;③in ethanol concentration range of 30% to 80% within the proportional division method to the same indicators and evaluation methods Rhizoma Cyperi is optimized by the residue after SFE-CO2 mention alcohol molecular weight< 1000Da Shen extract prepared a better alcohol concentration;④the same indicators and evaluation methods for the residue 5 ways (SBE method, WE method, AE method, SBAE method, WAE method) molecular weight< 1000Da extract more and more index components;⑤not necessarily due to the chemical equivalent of bioequivalence, pretending to be the dregs of 5 molecular weight≤1000Da extraction method extracts the main efficacy and Toxicity;⑥on Rhizoma Cyperi by SFE-CO2 extraction residue after 5 molecular weight≤1000Da extract and rabbit serum with HPLC fingerprint for Comparison;⑦extraction methods on the five kinds of molecular weight≤1000Da extract of rabbit serum containing rabbit intestinal smooth muscle for the role of comparison;⑧comprehensive evaluation of the final, optimized Rhizoma Cyperi by SFE-CO2 extraction residue after the best methods and process conditions.Results:①Rhizoma Cyperi meet the "Chinese Pharmacopoeia" under the 2005 version of one of the requirements for qualified slices;②Preparation of optimized molecular weight< 1000Da SBE extract conditions were:3 fried water pH were: 2.0,6.5,9.0; extraction time were:2.0,1.0,1.0 h;③optimized semi-bionic extraction optimal concentration of 60% ethanol precipitation, the best ethanol concentration of 55%;④Rhizoma Cyperi by the residue after SFE-CO2 5 molecular weight< 1000Da method extracts the value of the comprehensive evaluation index order:YSBE> YAE>YWE> YSBAE> YWAE;⑤main pharmacodynamic results show that the molecular weight of< 1000Da 5 ways to extract,SBE method to the most potent efficacy; acute toxicity test showed that the molecular weight< 10000Da 5 ways to extract the basic drug-free;⑥Rhizoma Cyperi residue after by SFE-CO2 5 ways to extract and the molecular weight of< 1 OOODa serum with HPLC fingerprint comparison shows that characteristic peaks of the total area to the largest group of SBE solution;⑦5 Different Methods for the molecular weight of≤1000Da extracts containing serum to serum containing SBE method intestine in vitro inhibition of the strongest;⑧comprehensive evaluation results show that, Rhizoma Cyperi by the residue after SFE-CO2 extraction is better to SBE.Conclusion:For the extraction of Rhizoma Cyperi, the first to SFE-CO2 extraction of volatile compounds, residue after extraction SBE then extracted by centrifugation-membrane filtration method extracts a molecular weight≤1000Da, its multi-index comprehensive evaluation best; SBE method< 1000Da extract and its metabolites and other extraction methods (AE method, WE method, SBAE method, WAE method)≤1000Da corresponding extract compared to SBE method extract high content of effective substance, the best pharmacological strong. Therefore, Rhizoma Cyperi after by SFE-CO2 extraction residue to SBE better.
