Objectives:To prepare PMS (premenstrual syndrome, PMS) rat model of liver-qi invasion permits to study the model of evaluation index; testing center in rats in different brain areas (hypothalamus, limbic lobe, frontal cortex and hippocampus) in ERa and ERP gene expression changes with premenstrual syndrome (PMS) and behavioral changes in qi inverse relationship. Extraction of brain tissue RNA using semi-quantitative RT-PCR method, the expression of ERa and ERpmRNA research, while the total protein in brain tissue using Western blot technique (western blot, WB) were detected in rat brain areas expression levels of ERa and ERβ.Results:In the non-acceptance period, as compared with the control group rats in model group rats the level of open-field test scores, vertical scores and total score were significantly reduced compared with model group, the level of open-field rats administered score, vertical scores and total score were significantly increased by using RT-PCR method showed that, compared with the normal group, model group and treatment group rat hypothalamus, limbic cortex and hippocampus of expression of ERαand ERβmRNA significantly lower (P<0.05), adjusted liver herbs Jingqianping granule intervention can change the expression levels of ERαand ERβmRNA reduced state. Western blot experiments show that, compared with the control group rats of model group rats frontal cortex, hippocampus, hypothalamus, limbic ERαand ERβexpression was markedly reduced (P<0.01); with the model group, treatment group of the brain District ERαand ERβexpression levels increased to varying degrees.Conclusion:Preparation of PMS electrical stimulation cage rat model of liver-qi invasion permits, open-field scores can effectively evaluate the model, the four brain areas in rats with ERαand ERβgene expression may reverse cards close to qi with PMS, suggesting that PMS irritability Inverse card may occur in the four brain areas with the abnormal expression of ERαand ERβmRNA higher correlation; At the same time, the Jingqianping granule can down-regulate the expression levels of ERαand ERβmRNA in different brain regions, which maybe one of the mechanism to treat PMS with liver-qi invasion.
