ObjectiveTo investigate clinical significance of human papillomavirus DNA testing and Thinprep liquid-based cytology in cervical disease;Whether HPV-DNA load can be as an indicator of severity of cervical disease;Discuss the relationship between HR-HPV infection and cervical erosion.Methods1. Cervical exfoliated cellular specimens of 167 patients were archived from cervical disease diagnosis and treatment center of the Department of Gynecology at the First Hospital of jilin University from August 2010 to March 2011. Patients were divided into normal control group, cervical erosion according to the examination. All cytological specimens exfoliated from cervix (collected by clinician) wereprepared using Thinprep liquid-based cytology test (TCT) system and Hybrid Capture II detect.Colposcopically biopsy endocervical curettage (ECC) were performed in the patients who had TCT≥ASCUS (atypical squamous cells of undetetemined significance) with positive results of high-risk HPV DNA or HPV-negative with TCT≥low grade squamous intraepithelial lesion (LSIL), then all biopsy specimen were examined by pathology. orrelation of HPV infectionResult1. The pathology results were used as gold standards, the positive predictive value TCT alone for≥CINⅠwere 64.71%(11/17),66.67%(20/30) for the HC2 test alone; 92.85%(13/14) for HC2 and TCT combination.2. HR-HPV positive rates in normal cervical was 27.39%(20/73),for the cervical erosion was 31.91%(30/94),there was no statistically significant between them(X2=0.35,P>0.05).3. HPV DNA mean levels were 51. 51 pg/ml in normal cervical, 39.08 pg/ml inⅠ°cervical erosion, for theⅡ°was 62.77 pg/ml,350.33 pg/ml inⅢ°, there was no statistically significant between them(P>0.05).Conclusion1. When HC2 - HPV DNA and TCT united detection of diagnosis, can improve the positie predictie of cervical highly lesions. 2. Cervical columnar epithelium ectopic not increase the chance of HPV infection.3. HPV DNA load was not increase with the cervical columnar ectopic.4. The joint test TCT and HPV DNA, the results are all negative, can lengthen appropriately screening interval.
