Breviscapine Combined With PNS Protection Of Acute Focal Cerebral Ischemia Studies In Rats

Objective:Through being associated Breviscapine and PNS observed in acute focal cerebral ischemia in rats intercellular adhesion molecule 1 (ICAM-1), 72h neuroethology neurological deficit score, infarct size and light microscopy changes in brain tissue, and to explore breviscapine PNS associated with acute cerebral ischemia induced brain protection.Methods: 50 rats were randomly divided into sham operation group, model group, breviscapine group, PNS group, traditional Chinese medicine combined with groups of 10, using suture model of middle cerebral artery occlusion. 2 hours after administration, once daily qd for 3 days. After each group were taken at 6h, 24h, 72h venous blood, 1 hour after the last blood brains were decapitated brain. Detected by ELAIS values of serum ICAM-1. Neurological deficit score using Bederson10 hours law standards, infarct area was calculated based on percentage. HE staining, electron microscopy observation of cellular pathology results.Results: Compared with the sham group, model group, serum levels of ICAM-1 was significantly increased, neurological deficit score high, large area cerebral infarction, cerebral cortex coronal sections shows a large majority of the globus pallidus infarction, and normal brain tissue (red) boundaries more obvious, some as deep as the caudate nucleus, the number of nerve cells around the area significantly reduced brain edema, showing the nucleus shrinkage or disappearance of vacuolization of neurons, nerve loose, pale staining, brain edema weight; Compared with model group, breviscapine group, PNS rats can be seen surrounding gliosis, lighter brain edema, ICAM-1 significantly decreased , neurologic deficits to a lesser degree, smaller cerebral infarction. And Compared with breviscapine group, PNS group, traditional Chinese medicine combined group was significantly glial cells proliferation, brain edema was significantly reduce, serum ICAM-1 significantly decreased neurological deficits lighter, smaller infarct; (P all <0.01).Conclusion: Breviscapine group, PNS, and combined with traditional Chinese medicine group, can reduce cerebral ischemia induced ICAM-1 values, so that significantly reduced neurological deficits, infarct size was significantly reduced, whereas traditional Chinese combined group be superior to Breviscapine Study group, PNS group.
