AIM:Research on transplantation of Mesenchymal stem cell (MSCs) of bone marrow to experimental emphysema rats, so as to provide scientific evidences for new therapeutic ways to chronic obstructive pulmonary disease.Methods:(1) 42 Wistar male rats were randomized into seven groups,which is group of control, groups of model (model 1w,2w and 4w group), groups of MSCs transplant (1w,2w and 4w group).3 rats come every group.(2)In 1-90 days, Wistar rats will be smoked with cigarette of Da qian men in the box twice a day.(40 minites each time, two intervals of 8 hours continues 6 days and rest one day)(3) Increasing MSCs of rat was isolated and cultured on method of adherence selection by a 6 week old Wistar rat. The cell vitality will be observed by Trypan blue staining.(4) All rats were killed after smoked 90 days. Separation lung tissue, fixing by paraformaldehyde after rinse lung cavity by physiological saline, the lower right lung lobewas paraffin embedding, leaves liced, HE dyed. Micro-computer image processing system was applied each slice choose 2 vision (x 200), mean alveoil number (MAN) and pulmonary alveolar area (PAA) than total.(5)Statistical analysis adopts SPSS13.0 processing data, seven groups of PAA between MAN and compares the single factor analysis of variance, compares the LSD t test opposite, inspection standards a=0.05.Results:(1) Model group and MSCs transplant group all appear emphysema pathological changes.(2) Model group rats activity decreases, fur noontide, body small and breathing fast, resting, panting, spiritual statuses has, MSCs transplant rats were obviously better than the model overall situation.(3) Model group lung volume enlarged, flexibility is poorer, a pale colour, the finger pressure can be left after dimpling, lungs cut more than the visible large quantities of alveolar space, present irregular shape, and size. Compare with Model group, MSCs transplant group lung lesions is lighter.(4) MSCs transplant group compared with model group:alveolar populations, the alveolar area is reduced, the alveolar septal thickening, transplantation group and model control group PAA mans, and comparative P> 0.05 was not statistically significant.Conclusion:(1) Mesenchymal stem cell transplantation can improve the whole condition, the most of lung specimen and the observation of pathology slices, and quota of pathology morphology study(MAN,PAA) of rats,which illustrare transplantation of MSC can reduce the lung lesions of obstructive pulmonary emphysema in rats.(2) The experiments after mesenchymal stem cell transplantation of rats compare pathology slices.The result is no statistically significance, indicating opportunity and proper time of experiment are pending further study.
