Objective: To study hMSC impact on the migration and proliferation of A-549 cells whichhas been conditioned by DDP and its mechanism.To observe the transition of MSCs by detectingthe expression of vascularendothelial growth factor(VEGF),transforming growth factor-β(TGF-β),fibroblast specific protein(FSP) and fibroblast activation protein(FAP),then discuss themechanism of MSC to repair the injury of tumor.Methods: hMSC and A-549 cells in mitotic phase were used to make hMSC conditionedmedium(MSC-CM) and A-549 conditioned medium(A-549-CM).hMSCs were conditioned for 2days in A-549-CM,then TGF-β1 and VEGF secretion was measured by enzymes linkedimmunosorbant assay(ELIZA).The long term conditioned MSC were conditioned withA-549-CM for 16 days.Cells were then analyzed by immunocytochemistry for the presence ofFAP and FSP expression.MTT method were used to detect the role of MSC-CM playing in themigration and proliferation of A-549 cells.Results: ELIZA showed that after 2 days conditioning by A-549-CM,MSCs have moresecretion of VEGF and TGF-βthan controls cells.Immunohistochemical S-P method showedthat after a long term conditioning,FAP and FSP were detected on MSCs for sure. MTT showedthat MSC-CM had evidently effects on the proliferation of A-549 cells.A-549 cells which wasconditioned by MSC-CM with DDP had a higher proliferation rate than that was conditioned byH-DMEM with DDP,and there were statistical significance(P<0.05)Conclusion: The reciprocal effect of MSCs and A-549 cells can increase the secretion ofVEGF and TGF-βof both.After a long term condition,MSCs can be transitioned to fibroblastlike cells.MSCs showed a positive effect to A-549 cells when it was injured by DDP.This maybecause of their tissue maintenance,repair and regenerative ability.
