ObjectiveTo investigate the effect of passive smoking on the expressional levels of histonedeacetylase2(HDAC2) in sensitized rats airway and interleukin-8(IL-8) in serum and theintervention of budesonide, explore the mechanism of airway inflammation of sensitized ratsaffected by passive smoking and the possible role of HDAC2 in the anti-inflammation effect ofbudesonide in airway.Methods50 healthy male Wistar rats were divided randomly into control group, sensitized group,smoking sensitized group, budesonid sensitized group and budesonid smoking sensitized group.ten rats each group. The rats in sensitized group were sensitized with 10% ovalbumin andchallenged with 2% ovalbumin to establish the sensitized model. the control group gave salineinstead of ovalbumin. From the challenged, smoking sensitized group were smoked passively.From the challenged, budesonid sensitized group and budesonid smoking sensitized group wereinhaled budesonid. airway inflammation by HE staining, The expressions of HDAC2 proteinwere defected using immunohistochemistry and IL-8 in serum were defected using ELISA in allrats of five groups.Results1.The expression of HDAC2 protein of the sensitized group(0.417±0.044)were lower thanthe control group(0.472±0.047)(P<0.05), smoking sensitized group(0.339±0.050)were lowerthan the sensitized group(P<0.05). The expression of HDAC2 protein of budesonid sensitizedgroup(0.462±0.021)were higher than those of the sensitized group(P<0.05). The budesonidsmoking sensitized group(0.425±0.035)were higher than those of the smoking sensitizedgroup(P<0.05). Compared with control group, The expression of HDAC2 protein were lower inbudesonid sensitized group, but the P values was more than 0.05, the expression of HDAC2protein were lower in budesonid smoking sensitized group, the P values was less than 0.05.2.The level of IL-8 of the sensitized group[(55.97±7.09)pg/ml]were higher than the controlgroup[(47.25±4.94)pg/ml], smoking sensitized group[(63.04±7.06)pg/ml]were higher than thesensitized group(P<0.05). The level of IL-8 of budesonid sensitized group[(48.92±4.51)pg/ml]were lower than those of the sensitized group(P<0.05), the level of IL-8 of budesonid smokingsensitized group[(54.32±7.62)pg/ml]were lower than those of the smoking sensitized group(P<0.05).Compared with control group, the level of IL-8 were higher in budesonid sensitized group, but the P values was more than 0.05, the level of IL-8 were higher in budesonid smokingsensitized, the P values was less than 0.05.3.The expression of HDAC2 protein were negatively correlated with the level ofIL-8(r=-0.548, P<0.05).Conclusions1.Passive smoking can aggravate sensitized rats airway inflammation by decreasing theexpression level of HDAC2 protein in airway and increasing the level of IL-8 in serum.2.Budesonid may suppress the inflammation of airway by increasing the expression level ofHDAC2 protein in airway.3.Smoking can weaken the anti-inflammatory effect of budesonid by influence the HDAC2protein...
