OBJECTIVETo develop the Rubrus Suarissimus S.Lee dental care buccal tablets(RSL) mainly composed of RSL extract and assess the effects of Rubrus Suarissimus S.Lee dental care buccal tablets(RSL) on adhesion and acidogenieity of S.mutans, in order to explore its anticariogenic activities and lay a foundation for the development and exploitation of Guangxi's special local plant RSL on prevention of dental caries in the future.METHODS1. Preparation of RSL dental care buccal tabletsSelect raw material in accordance with formula principles of health care food and dental health target, and choose RSL solution with its minimum inhibitory concentration (MIC) value lower than the MIC value determined by preliminary experiments for this subject as the added concentration of the RSL saponin in the buccal tablets. With the reference prescription provided by our subject team, the best prescription for the RSL dental care buccal tablets is determined through orthogonal experiments. Then use the traditional wet granulation methods to produce the tablets.2. Bacterial experiments of RSL dental care buccal tablets2.1 Oversaturation RSL dental care buccal tablets suspension and bacterial preparation2.1.1 The RSL dental care buccal tablets were ground into powder under aseptic condition and diluted into TTY liquid medium by two-fold dilution, of which concentration ranges from 5g/50ml to 0.313g/50ml.2.1.2 The freeze-dried S.mutans powder were cultivated in TSB for 3h→cultivated in TTY for 48h under anaerobic condition(37℃)→Colonies were centrifuged, picked and washed with 15ml sterilized normal sodium solution→adapted the optical density (OD) at 540nm to 1.2.1.3 The Inhibitory Effect of RSL dental care buccal tablets on adherence of S.mutansThe RSL dental care buccal tablets suspension was diluted by two-fold dilution with 1% sucrose TTY into five different concentration→added identical bacteria→inserted glass rod into the test tube, placed the test tube upright, anaerobic cultivated under 37°C for 48h→washed the bacteria adhered on the glass rod with NaOH solution→measured OD at 540nm→calculated the OD of bacteria adhered on the glass rod as rate of bacterial adherence.2.1.4 The Effect of RSL dental care buccal tablets on acidogenicity of S.mutans The RSL dental care buccal tablets suspension was diluted by two-fold dilution with 1% sucrose TTY into five different concentration→added identical bacteria→anaerobic cultivated under 37°C for 48h→calculated the pH value before and after the cultivation and change value△pH.RESULT1. A better prescription of RSL dental care buccal tablets is, each 100g RSL dental care buccal tablets is comprised of 5% RSL saponin, 20% microcrystalline cellulose and 74.5% dextrin 74.5g. The RSL dental care buccal tablets are double convex round tablet in light yellow green color. The outside surface is smooth with no split and visible impurities. It tastes cool and sweet, with a special flavor of Rubrus Suarissimus S.Lee.2. In adhesion experiments, OD value of S.mutans adhered on the glass rod gradually decreased with the increasing concentration of RSL dental care buccal tablets. There was significant difference between OD number of RSL dental care buccal tablets groups and the control groups. Thus RSL dental care buccal tablets could inhibit the adhesion of S.mutans. 3. In acidogenieity experiments, the pH values of each concentration group all decreased compared with that before the acidogenieity experiment. Only when concentrations less than or equal to 0.313g/50ml, P> 0.05, the difference was not statistically significant. In other groups, P <0.01, the difference was statistically significant. There is relatively significant difference for△pH value between over saturated RSL dental care buccal tablets groups and negative control groups, thus RSL dental care buccal tablets could inhibit the acidogenicity of S.mutans.CONCLUSION1. The best prescription of RSL dental care buccal tablets: RSL saponin 5.5g, microcrystalline cellulose 20g, dextrin 74.5g, menthol 1g.2. RSL dental care buccal tablets could inhibit the adherence of S.mutans on the surface of glass rod significantly.3. RSL dental care buccal tablets could inhibit the acidogenieity of S.mutans.4. RSL dental care buccal tablets as a new type dental care food, has some promising future in the field of detal caries prevention.
