OBJECTS: We use the formalin pathological pain model in rabbits to study the influence of intravenous and epidural of sufentanil analgesia on rabbitβ-endorphin (β-EP) and oxygen free radical (OFR).METHODS: 18 healthy rabbits were randomly divided into three groups, intravenous analgesia group (group A, n=6), epidural analgesia group (group B, n=6) and the control group (group C, n=6) . The rabbits in each group were injected 5% formalin 0.5ml to make formalin pathological pain model. Group A were injected sufentanil 0.46μg/kg by intravenous and group B were injected sufentanil 0.64μg/kg by epidural method for analgesia respectively. Group C was injected saline 0.5mL by intravenous as control. The spontaneous noticeptive behavior was measured by counting to assess the pain of animals, pain response was recorded with pain grade and two phase cumulative pain grade (the first phase is from 0 to 10 minutes and the second phase is from 11 to 45 minutes) for a period of 45 minutes. We deaw-off ear dege's venous blood before the experiment (T0), 10 minutes after analgesia (T1), 20 minutes after analgesia (T2), 40 minutes after analgesia (T3), 2 hours after analgesia (T4), 8 hours after analgesia (T5), 24 hours after analgesia (T6), 48 hours after analgesia (T7). The level ofβ-EP was detected by ELISA and OFR was by Maleic dialdehyde (MDA).RESULTS: Sufentanil analgesia can reduce the pain of rabbits and decrease the pain score of group A and group B steadily , and the pain of rabbits in group C last longer than that of group A and group B. The pain responses in both phase were inhibited significantly in groupA and groupB compared with group C (P<0.05). There was no significant difference in the pain responses of group A and group B during the two phase. The levels ofβ-EP and MDA in three groups had no significant difference before experiment. The level ofβ-EP in group A and B rose slightly compared with T0. But compared with group C, the level ofβ-EP in group A and B declined significantly. There was no significant difference in group A and B. The level ofβ-EP in group C showed a significant rose at T1-T7 compared with T0. The level of MDA dilined significantly in group A and B at T1-T7 compared with group C and T0 (P<0.05). The level of MDA rose significantly in group C at T1-T7 compared with T0. The level of MDA dilined in group B at T6-T7 compared with group A.CONCLUSION: The level of rabbitβ-EP and MDA were released by pain and stress. Intravenous and epidural injection of sufentanil analgesia can effectively controlβ-endorphin and MDA, and keep the internal environment of organism ,so as to reduce the pain reactions. The effect of epidural method is better than that of intravenous method.
