Establishing Tca8113 Adriamycin Drug-Resistant Tongue Cancer Cell Line And The Expression Of ADAM15 In TCA8113/ADM Cell

Abjective: To detect the expression level of Adamalysin15(ADAM15) of Tca8113 adriamycin drug-resistant tongue cancer cell line(Tca8113/ADM ) of tongue cancer and Tca8113 parental cell line after establishing Tca8113/ADM cell line of tongue cancer.Methods:1,Tca8113/ADM cell line was established by the method of drug inducing continually and the drug concentration gradient in 15 monthds . 2,the expression level of ADAM15 of Tca8113/ADM cell line of tongue cancer and Tca8113 parental cell line was detected by Western blotting, and then the relationship between the expression of ADAM15 in Tca8113 drug-resistant of tongue cancer and the oral cancer metastasis was analysed.Results:1,Tca8113/ADM cells growed steadily in the 1.2ug/ml ADM culture solution after 15 months. The resistant index of Tca8113/ADM cell line was 22.61. In the light microscope ,the soma of Tca8113/ADM cell was bigger and rounder than that of perantal cell, and cell nucleus and shape was irregular. There were more particles in Tca8113/ADM cells, which doubling time prolonged. 2,Compared with The expression of ADAM15 in Tca8113 parental cells, Tca8113/ADM cells was higher(p<0.01). Conclusions:1,the drug-resistent Tca8113/ADM cell line of tongue cancer is established successfully with the method of drug inducing continually and the drug concentration gradient .2,due to ADAM15 is an important biomarker of tumor metastasis, and its expression in Tca8113/ADM cell line is higher than perantal cell line. It indicates that the metastasis potential ability of Tca8113/ADM cell line may be stronger than the Tca8113 perantal cell line.
