According to the basic pathogenesis of HMG "Liver and kidney deficiencies, disorders of Chong and Ren; Qi stagnation, Phlegm, blood stasis as its Root Cause & Symptoms",the famous doctor of academic institution of TCM in sichuan province developed a related formula-Qingxiangrukang Particle which has efficacy of regulating qi-flowing and dissipating phlegm,softening and resolving hard mass, removing blood stasia for rlieving pain,which is used for treating HGM(for syndromes of stagnation of liver qi,intermingled phlegm and blood stais).This formula is designed by combining syndrome differentiation with modern experimental research and based on clinic observation for about 10 years, which showed its curative effect on preventing and curing mammary hyperplasia diseas,such as improving mammary hyperplasia diseas s breast pathological tissue damage and regulating hormone levels.Objective:To study on pharmacodynamics of QingXiangRuKang Particle which is used for Mammary hyperplasia disease with the model of whole animal experiment,it aimed to explore and further clarify its mechanism of action,and provide scientific basis for rational clinical drug-use hopfully.Methods:This subject mainly observed pharmacodynamic of QingXiangRu Kang Particle treating Mammary hyperplasia disease by several aspects as follo wing:1) Test On experimental rat model of mammary gland hyperplasia. Exper imental hyperplasia of mammary gland rats was induced by injection of estradi ol benzoate and continuously progesterone to the animal.After giving QingXian gRuKang Particle lasted for 30 days, the diameter of the second nipple of rats was measured before and after administration. The levels of serum E2 and ser um P were determined. The pathological changes of breast tissue was observed under microscope.The expression of ER,PR in breast tissue were observed by immunohistochemical technology. Study the efficacy and possible mechanism o f QingXiangRuKang Particle on experimental mastoplasia rat.2) Test On experi mental rabbit model of mammary gland hyperplasia. Experimental hyperplasia o f mammary gland rabbit was induced by injection of estradiol and progesterone to the animal continuously.Investigated whether Qingxiangrukang Particle has t he treatment of breast hyperplasia.3)study prevention of precancerous lesions of experimental breast cancer in rats induced by DMBA.Establish rat model of p recancerous lesion of experimental breast cancer by fed DMBA. Investigate wh ether Qingxiangrukang Particles has the prevention and treatment of precancero us lesion of breast cancer.4) effect of blood-activiating and stasis-dissolving.By examing the time of coagulation, the influence of tail thrombosis which carrage en induced and blood rheology in acute stasis model rats.5) Test anti-inflammat ory. By the models of granuloma induced with cott on ball in mice and xylen e-induced ear edema in mice, investigate anti-inflammatory effects of Qingxiang rukang Particle.6) Analgesic experiments. The hot plate method to induce pain and ip injection of acetic acid to establish mouse writhing model were used fo r examining the analgesic effect of Qingxiangrukang Particle.Results:1) The content of serum estrogen was increased (p<0.05),and microscopic observation showed marked hyperplasia of breast tissue of model g roup.The expression of ER,PR was was significantly increased. This showed t hat the model was successful. We found breast diameter was reduced after trea tmentthat of QingXiangRuKang particle, and hyperplasia degree of breast tissue was lightened. The given Qingxiangrukang Particle.42g crude drug/kg group can lower the content of serum estrogen. Qingxiangrukang Particle in three dos age groups can reduce the expression of ER,PR.2) The results show that Qing XiangRuKang particle has obvious therapeutic effect on experimental rabbit mo del of mammary gland hyperplasia.3) The results show that QingXiangRuKang particle has no significant inhibitory effect on of precancerous lesions of exper imental breast cancer in rats induced by DMBA.4) Three dosage groups can p rolong the time of coagulation. At the same time, they can inhibit tail thrombo sis induced by carrageen,and improve blood rheology in acute stasis model rats. 5) Experimental anti-inflammatory, three dosage groups can inhibit granuloma i nduced with cott on ball in mice and xylene-induced ear edema in mice.6) An algesic experiments. Three dosage groups have analgesic effect of pain induced by heat stimulation and chemical stimulation to a certain extent.Conclusion:QingXiangRuKang Particle has clearly effect for HGM in animal experiment,it may be related to its regulation of serum hormone and impact of ER, PR expression mechanism. Additionally. Also it has some improvement in blood rheology.
