Basic Experimental Study Of The Method Of Traditional Chinese Medicine "Huo Xue Hua Yu" On The Healing Of Bone Fracture

Objective:To explore the influence and mechanism of the method of traditional Chinese medicine "huo xue hua yu" on rat osteoblast function in vitro. To provide the basis for the method of traditional Chinese medicine "huo xue hua yu" on bone healing.Method:Select Fu Yuan Huo Xue Tang containing serum, the representative of traditional Chinese medicine "huo xue hua yu" for drug serum group, blank rat serum as a blank serum group, individual observation indicators with 10% fetal bovine serum as the control serum group. Serum pharmacology was applied to the investigation. Osteoblast cell was isolated from calvaria of newborn SD rats by means of modified sequential collagenase digestion and incubated in DMEM medium containing 10% fetal bovine. The cell morphology was observed under a phase contrast inverted microscope. MTT assay, ALP activity, AKP quantitative determination, mineral node count were carried out to determine the status and activities of proliferation and differentiation. To explore the influence and mechanism of the method of traditional Chinese medicine "huo xue hua yu" on rat osteoblast function in vitro through the observation of the indicators.Results:There's obvious difference in the three groups on MTT assay,and the proliferation of drug serum group is more significant than blank serum group and control serum group. There is no obvious difference in the two groups of mice serum in the short term on ALP activity, but obvious difference in the groups of mice serum and fetal bovine serum. On the mineral node count, all the two groups of mice serum increased the number of mineral nodes after cultured 15 days, and the drug serum group is more significant from the 7th day. (P< 0.05). On the AKP quantitative analysis the drug serum group can effectively promote osteoblast secrete alkaline phosphatase compared with blank serum group.(p< 0.05)Conclusion:Fu Yuan Huo Xue Tang-containing serum can promote rat osteoblast function in vitro. It provide the basis for the method of traditional Chinese medicine "huo xue hua yu" on promoting osteoblast function of bone fracture.
