PurposeTo observe the effect of Yuan Shi Sheng Mai Cheng Gu tablets played in differential and proliferation of MSC toward to osteoblast as well as MSC apoptosis, which aimed to providing theory evidence for ANFH.MethodsYuan Shi Sheng Mai Cheng Gu tablets were intragastric administered in adult SD rats for4days and drug serum was collected and isolated. We applied modified graded density centrifugation combined with adherent screen methods to extract SD rats MSC and expand to the third generation. We set4groups randomly, which were the control group (contained DMEM90ml, FBS10ml of L-DMEM culture solution), dexamethasone group (added10-7dexamethasone on the basis of the control group), drug serum group (added10%Yuan Shi Sheng Mai Cheng Gu tablets drug serum on the basis of the control group), and drug serum intervention group (adds10-7dexamethasone on the basis of the drug serum group). We measured the OD values in each group every day for5days, and drew the induced cells proliferation curve to present the effect on the proliferation of MSC in each group. Take the3rd generation BMSCs were randomly divided into2groups:drug serum group, traditional osteogenic group.When induced process completed, we observed BMSCs calcium condition through alkaline phosphatase(ALP) staining and alizarin red staining by biological inverted microscope.ResultsModified graded density centrifugation combined with adherent screen methods can achieve SD mice BMSC conveniently, economic, and effectively. Compared to the control group, the OD value in the drug serum group revealed high(p<0.05), which represented Yuan Shi Sheng Mai Cheng Gu tablets serum can improve BMSC proliferation.OD values in the dexamethasone group decreased and showed dexamethasone can inhibit BMSC growth and accelerate its apoptosis. OD values in the drug serum intervention group rising rapidly in the first three days, four days stabilized, began to decline in the first five days, compared with the dexamethasone group, the first day was no significant difference (P>0.05), the first2-5days there was a significant difference (P<0.05), decreased to a lesser extent, Yuan Sheng Mai Show marrow serum containing a piece of bone dexamethasone-induced mesenchymal stem cell proliferation and apoptosis have some protection effect. Both ALP staining and alizarin red staining results were positive in the drug serum group represented that Yuan Shi Sheng Mai Cheng Gu tablets serum can induce MSC toward osteoblast differential and proliferation.ConclusionYuan Shi Sheng Mai Cheng Gu tablets serum can improve BMSC proliferation and induce its differential toward osteoblast. It protected osteoblast from dexamethasone damage. And we supposed it may be the action mechanisms of ANFH. We suggested it can be used in tissue engineering because of its excellent osteocyte in vitro inducer.
