Objective By detecting the expression of cyclooxygenase-2(COX-2) and Human Eqidermal-growth- factor Receptor-2 (HER-2) in colorectal carcinoma patients, The paper probes into the relationships between COX-2 and HER-2, explores the correlations of them with clinicopathologic factors and effects of them to prognosis. Method Collecting the colorectal biopsy specinzens of 522 colorectal carcinoma patients hospitalized in First Affiliated Hospital of Anhui Medical Universit through 2005 Dec to 2007 Dec, the author performed expression detection of COX-2 and HER-2 by immunohistochemical technique, recorded clinicopathologic characteristics and followed up, calculated the survival time, analyed the relationships between COX-2 and HER-2, explored the correlations of them with clinicopathologic factors and effects of them to prognosis.Results 1) The positivity rates of COX-2 and HER-2 in colorectal carcinoma were 81.99% and 61.30% respectively. A significant correlation was found between the expressions of COX-2 and expressions of HER-2 (P<0.001). 2) No significant correlations between COX-2 expression and sex, age, site, differentiate degree, distance metastasis(P>0.05),but there were significant correlations between the COX-2 expression and tumor size, depth of invasion, Duke's stage,and lymph node metastasis(P<0.05). 3) No significant correlations between HER-2 expression and sex, age, site, differentiate degree, distance metastasis(P>0.05),but there were significant correlations between the HER-2 expression and tumor size, depth of invasion, Duke's stage,and lymph node metastasis (P<0.05). 4)The survival time were decreased in COX-2 and HER-2/neu expression positive patient(P<0.05).Conclusion COX-2 and HER-2 were high expression in colorectal carcinoma, and there were positive correlation between the expressions of COX-2 and expressions of HER-2. There were significant correlations between the COX-2 and HER-2 expression and tumor size, depth of invasion, Duke's stage, and lymph node metastasis. Expression of COX-2 and HER-2/neu were correlation with poor prognosis. COX-2 and HER-2/neu may be important biological makers for reflecting the invasion and metastasis of human colorectal carcinoma.
