Objective:(1) Explore the surgical method of interventional established rat model of sick sinus syndrome;(2) Comparison of formaldehyde, sodium hydroxide, acetic acid, ethanol, javanica oil which the five drugs best for the interventional rat model of sick sinus syndrome;(3) The experiment in comparative study of various indicators that intervention technique in rats with sick sinus superior model than thoracotomy method.Method:First ECG screening for healthy rats, selected 60 rats that heart rate between 350-450 beats/min, then randomly divided into six groups,they are formaldehyde thoracotomy group,formaldehyde in the intervention group, sodium hydroxide in the intervention group, glacial acetic acid in the intervention group, ethanol in the intervention group and intervention groups of Brucea javanica oil,10 each group. Formaldehyde thoracotomy group:conventional urethane anesthesia, connect ventilator, thoracotomy revealed sinus node, with 40% formaldehyde cotton ball wet compress sinus node area;Formaldehyde in the intervention group:anhydrous ether anesthesia, first the right edge of the second rat intercostal chest sternum cut a small mouth, the self mapping catheter along the insertion of this small chest, ECG waveform to determine the dose according to site,40% formaldehyde solution made with a micro-syringe mapping catheter by local infusion of modeling,other experimental methods in the intervention group with formdehyde in the intervention group. Measuring HR, pathological observation, recording and operation process and the survival rate after surgery were compared.Results:(1) Comparison with thoracotomy process involved in short spells, simple operation and low mortality rates;(2) The rats were decreased heart rate, heart rate changes through statistical analysis, obtained 10% sodium hydroxide as the most appropriate modeling of drug effect is stable;(3) From the histological analysis of view, ethanol, acetic acid, javanica oil although there is some damage to the sinus node, but the injury less severe; Formaldehyde and sodium hydroxide on the damage caused by sinus node closer to the clinical pathological changes of sinus disease.Conclusions:(1) Intervention method in rats with sick sinus model is simple, accurate positioning, vice damage, success rate.(2) Intervention rat model of sick sinus best test kit is 10% sodium hydroxide solution, their model has good stability, can be used for a longer period pharmacodynamics.
