| Objective: To observe the effects of butylphthalide on learning, memory and expressions ofβ-amyloid protein (Aβ) and NR2B in AD rat models'hippocampus. And to explore the possible mechanism of impact of butylphthalide on learning and memory in AD rats models.Methods: 32 rats were randomly divided into control group, model group and low dose, high dose of butylphthalide group, and every group had 10 rats. Aggregated Aβ1-40 was injected into the hippocampus of rats by stereotaxic coordinates method to induced Alzheimer's models. Aggregated Aβ1-40 was injected into each side of the hippocampus of model group and treatment group rats by micro syringes; the control group injected with saline. treatment group was lavaged with butylphthalide, and control group, model group were lavaged the same dose of vegetable oil. After two weeks, behavior was tested by the Morris water. Full four weeks after lavage rats were killed, and Aβin hippocampus , NR2B in hippocampal CA1 area was measured by immunohistochemical staining.Results: 1. The Morris water maze test results: Compared with the control group, model group rats had longer escape latency(P<0.01)and lower the percentage of time of crossing the platform(P<0.01), and both differences had the obvious statistically significant; Treatment group rats had significantly shorter escape latency (P<0.01)and higher the percentage of time of crossing the platform than model group(P<0.01), and both differences have the obvious statistically significant; the high dose butylphthalide of group than the low dose group improved in escape latency and the percentage of time of crossing the platform, but they had no statistical significance.2. The result of expression of immunohistochemistry: We had no see positive expression of Aβin hippocampus of control rats, and we found significant positive expression of Aβin hippocampus of model rats, and compared with the control group, model group rats had a significant difference in average gray value(P<0.01); Treatment group rats had significantly reduced in expression of Aβin hippocampus than control group, and the difference of average gray value was significant(P<0.01); NR2B in hippocampal CA1 area was only a small amount of expression in model group rats, and compared with the model group, the control group, treatment group was significantly NR2B expression , and the difference of average gray value had the statistically significant (P<0.01);expression of NR2B in the low dose group rats was less than the control group and the average gray value of the difference was significan(tP<0.05), but the high dose group and control group has no significant difference in the expression of NR2B.Conclusion: 1. Butylphthalide can improve spatial learning and memory of AD model rats.2. Butylphthalide can reduce the deposition of Aβof AD model rats'hippocampus, and promote the expression of NR2B protein in hippocampal CA1 area.3 The change of Aβand NR2b in hippocampus was related to the learning and memory of AD model rats. Butylphthalide maybe reduce the deposition of Aβin hippocampus and promote the expression of NR2b in CA1 area to improve the learning and memory ability of AD model rats. |
