Esophageal cancer is a common malignant tumor, With the very high morbidity and mortality,it is a serious threat to people's health. Surgery is the preferred treatment method, but many patients have lost opportunity when found, chemotherapy is still the mains treatment. Chemotherapy resistance is a major reason for Chemotherapy sustained and effective . The mechanism of tumor drug resistance is complex, In recent years, the increasing of DNA polymeraseβhave attracted more attention. To overcome drug resistance in addition to the development of new anti-cancer drugs,the search for resistance reversal agents is another direction. There are a lot of research to Western medicine , but because of its toxic side effects and the technological difficulties, high cost, are very limited in application. Chinese medicine because of low toxicity and reversal agents, is becoming the hot target.Objective:Actinidiae commonly used in clinical anti-tumor therapy ,So,its pharmacolon are being widlly concerned.The project studies Ec9706/cDDP cell line,uses modern medical techniques and quantitative indicators to observe whether plays a reversal of esophageal cancer chemotherapy resistance by reducing the expression of polβin esophageal cancer cells. This research hope seeks to suits the clinical use to bear the medicine reversal medicinal preparation.Methods:①with DDP(0.5μg/mL)in RPMI1640 medium was Ec9706/cDDP; using MTT cytotoxicity assay RAE to find non-toxic concentration of drug resistant reversal test; using MTT assay Ec9706/cDDP Kato pear with and without root extract cell survival after 48h culture, according to two sets of data with and without line of linear regression statistics were calculated for each group of half inhibition concentration (IC50), calculated reversal of the control group ratio = IC50 / test group IC50.②The cells were divided into experimental and control groups, the experimental group cells interfere with the RAE, RT-PCR were used to detect the expression of these two cell polβ.③Spss 13.0 software package were used to analyze the related data and contrast the result.the data to x±S Said, IC50 calculated using linear regression, compared two sets of data, using t test.Results:①RAE exist cytotoxicity on Ec9706/cDDP cell, and it is related to concentration of RAE.1,5,10μg/mL group and the control group showed no significant difference in inhibition (P > 0.05), Two inhibition of 100μg/ml group and 1mg/mlgroup were 18.3%,29.9%,compared with the control group,there was significant difference in inhibition (P<0.05).Therefore, the concentration of RAE 10μg/ml was selected as the best concentration toxic reversal experiments.②DDP alone, DDP on Ec9706/cDDP the IC50 for the 43.59μg/mL, DDP and non-small Cytotoxic dose 10μg/ml of RAE interaction,DDP on Ec9706/cDDP the IC50 decreased to 26.27μg/mL,the reversal fold was 1.66.③The control group of 10 samples in nine polβgene expression was high, and the remaining one showed a low level of expression; the experimental group of 10 samples in two polβgene expression was high,and the remaining eight were low the level of expression. Statistical analysis showed that polβgene in expression level in both groups there was significant difference (P<0.05).Conclusion:①RAE exist cell cytotoxicity on Ec9706/cDDP.②DDP alone,the IC50 of DDP on Ec9706/cDDP is 43.59μg/mL,DDP and non-cytotoxic agent interaction 10μg/mL the RAE, he IC50 of DDP on Ec9706/cDDP is 26.27μg/mL, the reversal fold was 1.66.It show that the RAE can be partially reversed Ec9706/cDDP resistant to cisplatin.③RT-PCR law examines two group of cell polβexpressions separately, the experimental group polβgene expression obviously reduces, showed that RAE may reduce in certain density scope Ec9706/cDDPpolβexpression level, this possibly is its reversal bears the medicine the mechanism, has provided the basis for it further clinical practice.
