Establishment Of Protein-marker Of Bleeding After Intrauterine Device Placement With Liver-depression And Blood-stasis And Study On Mechanism Of Bleeding

Objective:To find and identify protein-marker of bleeding after IUD placement with liver-depression and blood-stasis, and research molecular mechanism of bleeding of IUD placement with liver-depression and blood-stasis from serum protein level.Methods:From tracking and follow-up women who were IUD placement, the research collected a total of 310 serum samples before and after women who were IUD placement at different stages, which determined 85 efficacious. They were tested and analyzes by SELDI-TOF-MS. To find and identify protein-marker of bleeding after IUD placement with liver-depression and blood-stasis from compared protein map, and search empirical proteomic ontology knowledge base(EPO—KB) with analysis of Bioinformatics to find out their biological significance. Study on possible mechanism of bleeding of IUD placement with liver-depression and blood-stasis from diversifications of serum protein level.Results:1,Compare differences between women who were bleeding after IUD placement with liver-depression and blood-stasis and normal, then select 4 peaks(charge-to-mass ratios were 13797.1,6443.20,5934.95,3989.69). Establish decision tree model with 6 terminal nodes,then it were a accurate forecast for bleeding after IUD placement,and accuracy rate was 91.8% and sensitivity was 94.3% and specificity was 90.0%.2,Three serum proteins which charge-to-mass ratio were 3440.12.3949.87,5934.95 had differences in expression between women who were bleeding after IUD placement with liver-depression and blood-stasis and normal before and after IUD placement.That may be relate to constitution,and have important research value.3,Content of three serum proteins which charge-to-mass ratio were 7794.88.12870.7.13982.3 increased uniformly,but women who were bleeding after IUD placement with liver-depression and blood-stasis increased higher than the normal,however,may be directly relate to etiology.Conclusion:From inquiring protein database(EPO—KB), three serum proteins which charge-to-mass ratio were 3440.12,5934.95,7794.88 were neutrophil defensin 3 ND3. fibrinogen alpha chain Fgα,platelet factor 4 PF4.They were possible relate to mechanism of bleeding of IUD placement with liver-depression and blood-stasis,while Pointed out the direction for further research.The other serum proteins were not march,which may be imperfect protein database or experimental error.
