Effect Of Guanyin Mixture On Leptin And CCK Of Young Anorexia Rat

Objective:To observe the effect of Guanyin mixture on Gastrointestinal Hormone of rat model which suffering from anorexia and probe deeply into therapies and fundamentals of Guanyin mixture applied to child anorexia. Furthermore, it is farther expected to lay the foundation for developing new drug.Methods:Sixty healthy and grade SPF SD rats, days of age are 35～40 d, the weight were 63.81±7.95 g, male and female was fifty-fifty. The animals used in the laboratory were randomly separated into five groups, namely normal group,model group,masculine group(the group of Shenlingbaizhu mixture),the high-dose group of Guanyin mixture, each group contained twelve rats. The normal group was fed on conventional mash, the rest group simulated the main course of anorexia in child, adopting the tailor-made mash which made from Shouchuan Wang simulation method to fed the rats for three weeks, designing the young age rats model which suffering from anorexia, when the animals decreased significantly in food intake below the thirty to fifty percentage of the normal group, the weight was below the ten to fifteen percentage of the normal group, then the model was made successfully. The fourth week, each group was fed on conventional mash, and from then to, they began to treat and spend three weeks on them. After treatment, taking the blood from the abdominal main artery of the rats in each group, RLA using detection technology to measure the amount of the serum Leptin and the plasma CCK in rats.Results:The weight and food intake decreased obviously in every model group, the distinction have the statistic regularity which compare with the normal group(P<0.01); the content of serum Leptin in model group decreased, but the plasma increased, the distinction had statistic regularity which compare with the normal group(P< 0.05 or P<0.01). The concentration of serum Leptin which was treated by Guanyin mixture was higher than that in the model group, the distinction had no statistic regularity when the high-dose and the small-dose groups compared with the normal group (P<0.05), and the high-dose group was further accessed to the normal group. The concentration of the plasma CCK significantly increased compared with the normal group (P<0.01), the high-dose and the small-dose group of Guanyin mixture can both decrease the concentration of plasma CCK (P<0.01), but neither reach the normal level, the distinction between the two dose group had no statistic regularity.Conclusion:1. The Guanyin mixture can effectively improve the situation of the model rat's anorexia and increase the rat's appetite and food intake.2. It was primarily cognized that the mechanism of curing the model rats which suffering anorexia used Guanyin mixture were achieved by controlling the secretion of the outside Leptin and CCK.3. The high CCK concentration is one of the causes of anorexia.4. It was no cause of Leptin which excreted too much to lead to the appetite of the model rats which suffering from anorexia decreased; Decline in the level of serum Leptin is not the cause of anorexia, but rather the result of anorexia.5. The result of our research was to use traditional Chinese medicinal herbs to provide the theories and experimental data for further exploring the treatment of infantile anorexia mechanism; furthermore, it was expected to explore effective new Chinese medicinal herbs.
