ObjectiveTo detect the expression level of AEG-1 in human brain glioma, intracranial benign tumor and normal brain tissue, and investigate the association between the expression of AEG-1 and pathological grade(WHO I-IV) of glioma. Study provide the basis for gene diagnosis of glioma.MethodThe expression of MTDH in human brain glioma, intracranial benign tumor and normal brain tissue was detected by immunohistochemistry and the expression level of AEG-1 in human brain glioma, intracranial benign tumor and normal brain tissue was detected by Real time PCR. And then, the expression result statistics were analysed by SPSS 17.0.Results1. Immunohistochemical analysis showed that 93 of 102 paraffin-embedded glioma samples and 2 of 20 paraffin-embedded the intracranial benign tumor samples exhibited positive expression of MTDH. The others all exhibited negative expression of MTDH. The expression of MTDH was mainly localized in the cytoplasm.2. Real time PCR detection showed the normalized AEG-1 amount relative to normal brain(2-△△CT) was 1.78±1.43 in normal brain tissue, 8.25±7.23 in intracranial benign tumor,283.92±363.30 in glioma; Pathological grade:WHO Grade I:75.53±60.00, GradeⅡ:129.66±87.00, GradeⅢ:263.51±147.79, GradeⅣ:863.61±561.18.3. Statistical analysis suggested that MTDH and AEG-1 were significantly upregulated in primary human brain glioma compared with the intracranial benign tumor and normal brain tissue(P<0.05),and the expression of AEG-1 was significantly positive correlated with the pathological grade of glioma(WHO GradeⅠ-Ⅳ) (P<0.05).Conclusions1. The expression of MTDH and AEG-1 were significantly upregulated in primary human brain glioma compared with the intracranial benign tumor and normal brain tissue, AEG-1 might play a role during the development and progression of glioma.2. The expression level of AEG-1 was significantly positive correlated with the malignant degree of glioma.3. AEG-1 might become a novel, useful diagnostic and therapeutic marker for glioma.
