Objective To study the effect of CTGF on fracture healing. And search for the ways of CTGF application in the fields of bone tissue engineering and clinical fracture treatment. Effect of CTGF treatment on bone repair in a rabbits closed femur fractureMethods A 5 mm gap was performed in right mid tibials of 48 New Zealand white rabbits.The rabbits were divided randomly into two groups such as group A and B. The A group was injected subcutaneouly in CTGF 0.10ug/kg every other day for two weeks. The group B was injected 0.9%NaCl as the control group . we investigated the treatment. Effect of CTGF treatment on bone repair in a rabbits closed femur fracture, Modle using Radiological Bone density test, Blood phosphonium, calcium,Blood ALP,histomorphometric,Biomechanical analyses.Results X ray results demonstrated that two groups of bone defect callus formation was not obvious after two weeks, For four weeks: A group of defects nearly disappear, callus formation was more; B group of defects becomes vague, callus was less .Eight weeks: A Groups of callus was more, medullary cavity recanalization; B group of callus was less than A, medullary cavity did not pass. DEXA bone defect area can be seen that the results of the BMD and the BMD ratio was increased with the time of the same time, A group was increased significantly compared with the B group ,The difference was significant (P﹤0.05), A group of BMD has reached side of the same parts of the level after eight weeks, BMD increases in the ratio shows the same situation. Histological examination results of the experimental group of trabecular bone size, shape, arrangement basically normal, after 8 weeks,Control group: fibrous tissue and a large number of visible trabecular bone. Serum calcium,phosphorus and alkaline phosphatase content are significant differences of the experimental group and control group. Reverse experiments measured four parameters: torsional stiffness, maximum torque, maximum torque, angular displacement and reverse the failure in the next needed energy into the opposite side in the average a full percentage of the tibia after the parameter value, A were also much more than the group B when they were normalized as a percentage of the value for the contralateral, intact tibial. It has difference (P﹤0.05,P﹤0.01).Conclusions CTGF can encourage the fracture healing, to decrease soft tissue edema, to add bone density, and increased the biomechanical properties of a healing osteotomy site .
