Objective To study the effect of LIVIAL on osteoporosis association with femoral fracture in rats, and to provide the possibility of treatment for postmenopausal osteoporotic fracture.Method Fifty-four Sprague-Dawley(SD) female rats, aged 6-month-old, were divided into three group randomly: OVX group, SHAM group and LIVIAL group. The osteoporotic model was established by ovariectomy. The right femur was broken by operation at 8 weeks later. LIVIAL was given through gastromy, 0.26mg/kg/d, immediate after operation in the LIVIAL group. The rats in three groups were killed at the end of 2, 4, 6, 8 weeks postoperatively respectively for study. The study index include: the bone of X-rays, the histological of bone calluses (HE dye and Masson dye), immunohistochemistry of bone collagen I, the diaphyseal density of left femur, and the biomechanical properties of right femur.Results 1. The density of calluses was decreased obviously in OVX group. The line of fracture was still exist in OVX group while the line of fracture were unclear or disappeared in SHAM and LIVIAL group. 2. In the OVX group there was a little more cartilage during the early callus formation stage and fewer nunber of osteoblasts and more number of osteoclasts onsurface of osseous trabecula than other two groups. The osseous trabecula became thinner and disruption obviously in OVX group, and it became massive, thicker and closer gradually after 8 weeks postfracture. The area of osseous trabecula was bigger in the LIVIAL groyp than in the OVX group and SHAM group.3. The collagenous fibers was arranged looser and more irregular in OVX group than in SHAM and LIVIAL group. 4. The diaphyseal density of left femur was thicker in the LIVIAL group than in the OVX group after 8 weeks postfracture. 5. The consequence of elastic modulus, maximum stress, elastic deformation, destructive intension of right femur in LIVIAL group were the better than in the OVX group.Conclusion LIVIAL can make the quantity of bone increased gradually, the resorption of bone inhibited, the remodeling of bone accelerated, collagen of bone closed, biomechanics properties better. So LIVIAL was benefit to repair the fracture of bone in the late stage, and help postmenopausal fracture healing and prevent the next fracture.
