Background:At present, cardiovascular disease was principal harm to human heathy, expecially coronary heart disease, acute myocardial infarction (AMI) harm is the most serious, the mortality rate is extremely high. According to WHO report, acute coronary occlusion will become to 2020 the main cause of death in the world. To recover as soon as possible coronary occlusion which lead to schemia and hypoxia have the very vital significance in myocardial salvage. AMP579 as a new Adenosine artificial in recent years,compare to adenosine, has its unique function in suppress platelet aggregation, expanding coronary atery, diminution infarct size; but it has not been reported that they affect on CD62P.Objective:To explore two drugs the effect of the different doses adenosine and AMP579 to the expression of adhesion molecules CD62P on platelet in patients with coronary heart disease (CHD).Methods:The cubital vein blood of the patients and normal healthy individuals were collected, and were given different doses of adenosine and AMP579, the expression of CD62P on platelet of the patients and normal people were measured by enzyme-linked immunosorbent assays(ELISA).Results:Expression of CD62P in surrounding blood in patients with ACS was markedly higher than those in normal cases(p<0.05),and expression of CD62P in patients with those acute myocardial infarction(AMI)and unstable angina(UA)were significantly different(p<0.05). For Expression of CD62P in UA patients,there are statistical difference between using AMP579 groups and using adenosine groups (F=52.035,p<0.01); For AMI patients,there is significantly statistical difference between using AMP579 groups and using adenosine groups (F=68.837,p<0.00); For normal individuals,there is not statistical difference between using AMP579 groups and using adenosine groups(F=2.832,p>0.05). The half-logarithm line graphs see that the expression of CD62P degraded with the increasing concentrations of adenosine and AMP579 in UA and AMI groups. Compare to adenosine, in using AMP579 groups,the decreasing trend of expression level of CD62P was more obvious. Expression of CD62Pin surrounding blood in patients with AMI were total significantly different (F=85.632, p<0.01)with the different concentration of AMP579, with concentration of AMP579(450umol/L, 1000μmol/L,3000μmol/L) were not statistical difference(p>0.05),but they were statistical difference with Oμmol/L,150μmol/L(p<0.05). Expression of CD62P in patients serum with UA were total significantly different(F=135.735,p<0.01)with the different concentration of AMP579, with concentration of AMP579(450μmol/L, 1000μmol/L,3000μmol/L) were not statistical difference (p>0.05),but they were statistical difference with Oμmol/L,150umol/L(p<0.05). Expression of CD62P in patients serum with ACS were total significantly different (F=74.079,p<0.001)with the different concentration of adenosine, with concentration of adenosine (450μmol/L, 1000μmol/L,3000μmol/L) were not statistical difference, but they were statistical difference with Oμmol/L,10000μmol/L(p<0.05). The expression level of CD62P and CRP,APOB,TG,LDL-C and LP(a)was positive correlated,and the expressionlevel of CD62P,APOA and HDL-C was negative correlated.In patients with AMI,the expression level of CD62P,CK-MB and aTnl was positive correlated.Conclusions:Expression of CD62P in the ACS patients was markedly higher than those in normal cases.Expression of CD62P in patients with AMI was higher than in those with UA. It hinted that CD62P should be regarded a significant target in the occurrence of ACS. For UA patients,there is statistical difference between using AMP579 groups and using adenosine groups(p<0.05); For AMI patients,there is still statistical difference. By the data in the graph 1 and suggested that the lowest effective concentration of AMP579 is lower than adenosine,it was showed that AMP579 was superior adenosine to theplatelet surface CD62Pon the same concentration. It is favourable to make drug dosage in clinical application. Owing to the inhibitory action of CD62P expression enhance with the difference concentrations of adenosine and AMP579 (150μmol/L,450μmol/L, 1000μmol/L,3000μmol/L,10000μmol/L), it suggested that they enhance the role of inhibiting platelet activation. But two drugs were not difference with the difference concentrations(450μmol/L, 1000μmol/L,3000μmol/L), it hinted that it was possible to owe to receptor saturation.By related analyzing CD62P and blood-fat index, it suggested thatdecreasing CRP,APOB,TG,LDL-C,LP(a),CK-MB,aTnI and increasing APOA,HDL-C possibly decrease the expression level of CD62P in patients with ACS.
