Objective To study the function change of minocycline for the injury of retinal in rabbits with chronic ocular hypertension (OHT), for the drug used in ophthalmology treatment disease of optic nerve damage in the domestic clinical provide experimental basis for research.Methods Thirty rabbits were randomly divided into groupA(control group), groupB(model group) and groupC(drug treatment group),10 rabbits in each group. GroupB and groupC were made a chronic ocular hypertension (OHT) model on right eyes in each rabbit.The OHT models were made by injecting 0.3% carbomer 0.1ml into anterior chamber after take suction aqueous humor 0.1ml in rabbits of groupB and groupC. Group C was taked orally with 50mg minocycline (50mg*kg-1*d-1) for 30 days,the dose was doubled the first time.we measured and recordde ocular tension with Schiotz tonometer every week. Every group was detected optical coherence tomography(OCT),Flash electroretinogram(FERG) and Flash visual evoked potential(FVEP) on right eyes in each rabbit. We could observed thickness of layer of optic fibers with optical coherence tomography(OCT);We could observed the decrease degree of the amplitudes of the a-waves and the b-waves by FERG;We could observed the prolongation degree of the latent time of N1,P1,N2 of FVEP. At last, OLYMPUS optical microscope in the 10×40 times the microscopic observate the retinal layers of pathological changes.We analysised results statistically with SPSS13.0.Results The results of ocular tension showed:during the entire experiment, The ocular tension of groupB and groupC were higher than 22mmHg, the highest up to 55mmHg, and in the various time points after modeling were higher than the A group.The results of retinal OCT showed:GroupB had the thinest thickness of layer of optic fibers,the next one was groupC, the thickest one was groupA.There were significant difference in every two group(P<0.05). The Results of the FERGs showed:the amplitudes of the a-waves was no significant difference in each group(P>0.05).Compared to groupA, the amplitudes of the b-waves declined in groupB and groupC,groupB was obviously decliner than groupC,there were significant difference in every two group(P<0.05); The Results of the FVEPs showed:The latent time of peak was delayed in groupB and groupC,groupB had stronger inhibiting action on the latent time than groupC, there were significant difference in every two group(P<0.05). The results of Light microscope,showed:the retinal tissue layers of groupB was the most disordered,the retinal tissue layers of groupA was the most complete, Compare to groupA, groupC has structural changes in the retinal tissue layers, but lower than a slight change in groupB. Conclusion minocycline for the injury in the retina of rabbits with chronic ocular hypertension (OHT) had the protection functions.
