Objective In this study we investigated the mechanism of takeing minocycline is neuroprotective effect on chronic ocular hypertenstion models.Methods:48 rabbits were randomly divided into three groups:group A(control group), group B(chronic ocular hypertension),and groupC(Minocycline group).The right eye of chronic ocular hypertension and Minocycline group were used to build chronic ocular hypertension model by posterior chamber injection of 0.1ml of 3g/L Carbomer solution.The left eye were normal eye.After modeling, Minocycline group rabbits took Minocycline every day,50mg/kg respectively. The intraocular pressure(IOP) was monitored every week at the same time. The animals were sacrificed at 29 days after feed the medicine. The expression of caspase-3 in eight eyeball from every group was detected by immunohistochemistry,and the contents of malondiadehyde (MDA),superoxide dismutase(SOD) and NO in retina were assayed and compared among groups.Result:The IOP in B and C group increased and they had no obvious difference.The level of SOD in Group B and GroupC was markedly lower than that in group A(P<0.05),the content of MDA,NO and the IOD of caspase-3 were visibly higher than that in group A. The level of SOD in Group B was obviously lower than that in group C and the content of MDA,NO and the IOD of caspase-3 were considerable higher than that in group C.Besides, the level of SOD in Group C was significant lower than that in group A and the content of MDA,NO and the IOD of caspase-3 were markedly higher than that in group C.Conclusion:Minocycline is neuroprotective in chronic ocularhypertension model rabbits,It can protect retinal neurons by maintaining regulating production of SOD,MDA,NO and expression IOD of caspase-3 in chronic ocular hypertension model rabbits.
