Objective:To explore the pathogenesis of preeclampsia, the gene expression of IFN-y mRNA and IL-4 mRNA on the human placentas from patients with preeclampsia were detected by in situ hybridization method.Methods:20 cases of NLP women and 40 cases of patients with preeclampsia in the third trimester were observed in this study. The expression of Thl cytokines [interferony (IFN-y)] and Th2 cytokines [interleukin4(IL-4)] on the placenta were detected by in situ hybridization method. Furthermore,multi-function precise color-image measure system was used for quantitative analysis.Results:(1) The expression of IL-4 mRNA in normal pregnancy group (0.1870±0.0170), mild pre-eclampsia group (0.1810±0.0189) and severe pre-eclampsia group (0.1779±0.0190) decreased, Among the three groups, the expression of IL-4 mRNA was no statistical significance (P>0.05);(2) The expression of IFN-y mRNA in severe pre-eclampsia group (0.1799±0.0166) was significantly higher than those of normal pregnancy group (0.1614±0.0084) and mild pre-eclampsia group (0.1699±0.0100) (P<0.05); The expression of IFN-y mRNA in normal pregnancy group and mild pre-eclampsia group was no statistical significance(P> 0.05);(3) Compared with the ratio of IFN-ymRNA/IL-4 mRNA in normal pregnancy group (0.8687±0.0781), mild pre-eclampsia group (0.9458±0.0851) and severe pre-eclampsia group (1.0224±0.1412) increased significantly.Conclusions:The expression levels of Thl cytokines(IFN-y mRNA)those mostly play an immunologic injure role in pregnancy in severe pre-eclampsia group are higher than those in the other two groups, but Th2 cytokines (IL-4 mRNA) those mostly play an immune protection or nutritious role in pregnancy are indifference. So the high expression of IFN-γmRNA on the human placentas from patients with preeclampsia is related to the disease, and in the development of preeclampsia, the effect of IL-4 mRNA is not obvious; The increased ratio of IFN-ymRNA/IL-4 mRNA suggest that the Th1/Th2 cytokines imbalance in maternal2fetal interface is one of the pathogenesis of preeclampsia.
