BackgroundThe pathogenesis of preeclampsia is still unknown. Insufficient trophoblasts infiltration influences the physical vascularremoulding of spiral arterial. And the resistance of the terminal vessels in placenta increases,and the efficient placenta perfusion cannot be accordant of the development of pregnancy. Transforming growth factor-beta1(TGFβ1)and Matrix metalloproteinses-9(MMP-9) may lead to trophoblasts infiltration deficient and be involved in the pathogenesis of preeclampsia.ObjectivesStudy on expression of TGFβ1 and MMP-9 in the placenta tissues in order to evaluate the role of them and their relationship in preeclampsia.Methods:1,Immunohistochemical S-P method was used on the normal group(15 cases) and the preeclampsia group(24 cases in total)to observe the location and distribution and level.2,Reverse transcription polymerase chain reaction was used on the two groups to quantitatively determining TGFβ1 mRNA and MMP-9 mRNA in the placenta tissues. Results1,S-P method: The levels of TGFβ1 expression in the severe preeclampsia group were significantly higher than those of normal and the slight group(p<0.05),and the expression of TGFβ1 was no significant difference between the slight preeclampsia and normal women(p=0.087);The levels of MMP-9 expression in the severe and slight preeclampsia group were significantly lower than those of normal group(p<0.05).There was a significantly negative correlation between the expression of TGFβ1 and MMP-9 in placental tissue(p<0.05).2,RT-PCR: The levels of TGFβ1 expression of severe preeclampsia group were significantly higher than those of slight preeclampsia and normal group.The levels of MMP-9 expression of severe preeclampsia group were significantly lower than those of slight preeclampsia and normal group.There was a significantly negative correlation between the expression of TGFβ1 and MMP-9(p<0.05).Conclusion1,The abnormal expression of TGFβ1 and MMP-9 in placenta was reduced at the level of transcription and protein and leads to insufficient trophoblasts infiltration and might be involved in the pathogenesis of preeclampsia.2,There was a significantly negative correlation between the expression of TGFβ1 and MMP-9 in placental tissue and cued that they might commonly participate in the pathogenesis of preeclampsia.
