OBJECTIVE:To investigate the effect of chronic alcohol consumption on vascular responses of isolated aorta,blood pressure and anti-oxidation function in rats and taurine intervention.METHODS:Thirty male Sprague-Dawley rats were randomly assingned into 3 groups:control group,alcohol group and taurine group.The rats in the control group drink water,The rats in the alcohol group drink wine free for 5 months, In the taurine group,The rats were given taurine on the basis of drinking wine.We determine body weight and blood pressure every two weeks.The effect of the vasoactive substances on the pre-contraction induced by NE 10-5mol/L and KCl 60mmol/L were observed with MS4000 computer system.;Then we determine the content of SOD,MDA and GSH-Px in serum and the vital organs of each group.RESULTS:Taurine lower the body weight 8-16 weeks in taurine group compared with alcohol or control group(P<0.05).The SBP and DBP 12-20 weeks in alcohol group higher than control group;The SBP and DBP 12-20 weeks in taurine group lower than alcohol group(P<0.05). The relaxation effect of Pina(10-6mol/L) on the pre-contraction induced by 10μmol/L NE were higher in taurine group than alcohol group(P<0.05).The relaxation effect of Pina(10"8mol/L and 10-7mol/L) on the pre-contraction induced by 60mmol/L KCl were higher in taurine group than alcohol group(P<0.05).The relaxation effects of Amrinone(10-8mol/L 10-7mol/L and 10-6mol/L) on the pre-contraction induced by 10μmol/L NE were higher in taurine group than alcohol group(P<0.05).The relaxation effect of SNP(10-8mol/L) on the pre-contraction induced by 10μmol/L NE were higher in taurine group than alcohol group(P<0.05).The level of SOD and GSH-Px in serum,heart,liver kidney and brain of alcohol group were lower than control group. (P<0.05),The level of MDA were higher than control group (P<0.05).The level of SOD in serum,heart and liver of taurine group were higher than alcohol group(P<0.05),The level of GSH-Px in serum,heart,liver,kidney and brain of taurine group were higher than alcohol group(P<0.05), The level of MDA were lower than alcohol group(P<0.05).CONCLUSION:The vasodilative response of Pina,Amrinone and SNP in isolated aortic rings of rats with alcohol group did not change significantly. But can lead to hypertension and reduced anti-peroxidation of rats. Taurine can reduce the blood pressure by against lipid peroxidation and by enhancing the relaxion effect of vasoactive substances.
