Objective By investigating the transcriptional level of Bmi-1 in lung adenocarcinoma SPCA1 cell line and mRNA and protein level in human lung cancer tissue so as to explore the role of Bim-1 as a new marker in clinical diagnosis and prognosis evaluationMethod (1) RT-PCR were used to detect the mRNA expression level of Bmi-1 in lung adenocarcinoma SPCA1 cell line and NSCLC tissue and were compared with normal pulmonary epithelial cells and mucosa of normal bronchus. (2) Immunohistochemistry were applied to detect the protein expression of Bim-1 in 48 lung cancer tissues, 29 peficancerous tissues and 13 benign lesions of lung. The relation of Bim-1 expression and clinical features and pathological classification of lung cancer were analyzed.Results (1)The mRNA expression of Bmi-1 was significantly higher in human lung adenocarcinoma SPCA1 cell than in normal pulmonary epithelial cells;The mRNA expression levels of Bmi-1 in 20 NSCLC tissues were higher than normal mucosa of bronchus. (2)The protein expression level of Bmi-1 in NSCLC tissue was significantly higher than in lung benign disease and adjacent tissue (P<0.05), but no significant correlation is detected in lung benign disease and peficancerous tissues (P>0.05). (3) No significant correlation was detected in histological subtypes. (4)The protein expression levels of Bmi-1 in poorly differentiated lung cancer were higher than in well-differentiated lung cancer. (5)The protein expression level of Bmi-1 inâ…¢andâ…£stage lung cancer was higher than inâ… andâ…¡stage, but no significant correlation was detected inâ… andâ…¡stage lung cancer. (6) The protein expression levels of Bmi-1 were higher in NSCLC groups with lymph node metastasis than without lymph node metastasis.Conclusion The expression of Bmi-1 in NSCLC either in mRNA or protein level is significantly increased and is correlated with TNM staging and lymph node metastasis;Bim-1 can serve as a new marker for evaluating the extent and prognosis of NSCLC.
