ObjectiveTo investigate the expression of effect of HER2 on COX-2/PGE2/P450arom signal pathway in human endometrial carcinoma transplanted on nude mice.MethodsNude mice were transplated subcutaneously with Ishikawa cells, Ishikawa cells stably transfected with HER2 plasmid, Ishikawa cells transfected with empty plasmid, respectively. The tumor volume and weight were measured. Real-time PCR and Western blot were used to detect the levels of HER2,COX-2,P450arom in tumor tissues. The levels of PGE2,E2 were detected by ELISA.ResultsThe tumor formation rate and tumor size in HER2 plasmid transfection group was noted significantly higher than control group(P<0.05). After transfection, Expression of HER2 mRNA and COX-2 mRNA were significantly rised in HER2 plasmid transfection group compared with control. The protein levels of HER2,COX-2,P450arom were significantly higher in HER2 plasmid transfection group compared with control(P<0.05). Expression of PGE2,E2 were significantly higher in HER2 plasmid transfection group than those of control groups.ConclusionsHER2 promotes the growth of Ishikawa cells by regulating the COX-2/PGE2/P450arom pathway to affect the expession of E2 in vivo.
