Experimental Research Of Therapeutic Effects Of ZARY On Adjuvant-Induced Polyarthritis In Rats

The basic pathological changes of Rheumatoid Arthritis are chronic arthrosynovitis for the formation of pannus invasive, RA is a common chronic systcmic autoimmune disease. Rheumatoid arthritis can attack adult in any age. 80% of these come on 30~50 years old, but the incidence of RA in 60 years old was obviously higher than 30 years old. RA often attack slowly and insidiously, if RA can not be diagnosed and treated timely, 70% of RA patients will have severe side adverse effect after 2 years.Object: To observe the therapeutie effect of the ZARY on Ajuvant Athritis (AA) of rat and compared with hexadecadrol. This can provide experimental and theoretical basis for using of "ZARY" in treatment of rheumatoid arthritis.Method: The experimental uses modified Freund's complete adjuvant-induced model of adjuvant arthritis in rats. The model rats were given 0.1ml subcutaneous complete Freund's adjuvant on the left hindlimb. This experiment used different concentrations of drugs to cure the adjuvant arthritis in rats. In the experiment 1, we used preventive medication, that is, the first time medication was designed 1h before proinflammatory, and the rats in each group were separately given drugs by normal saline 0.05ml each rats, hexadecadrol 0.9ml/kg, ZARY 0.4mg/kg, 4mg/kg, 8mg/kg; the second time medication was designed after 7 days, and the rats in each group were separately given drugs by normal saline 0.05ml each rats, hexadecadrol 0.9ml/kg, ZARY 0.4mg/kg, 4mg/kg, 8mg/kg, we gave the medication for 7 days. In the experiment 2, the first time medication was designed after 15 days, the rats in each group were separately given drugs by normal saline 0.05ml each rats, hexadecadrol 0.9ml/kg; ZARY 0.4mg/kg, 4mg/kg, 8mg/kg, we gave the medication for 11 days.In order to evaluate the effect of the drugs and searching mechanism of action, some variances were measured at different time, such as the weight change, the swelling degree in lelt hind paws, the aural region macula and the tail tubercle etc.Result: ZARY could inhibit the primary swelling of the left paws of rats, indicating that ZARY can inhibition acute inflammatory response on the partial proinflammatory of AA primary phase; ZARY could inhibit the secondary swelling of the paws of rats, at the same time ZARY can improved the aural region macula and the tail of nodular lesions of secondary symptoms; besides thymus and spleen index indicates that ZARY has immune modulation with anti-inflammatory action in AA rats.Conclusions: ZARY has a good anti-inflammatory and immunomodulatory effects.
