Study On Quality Standard Of Extractum Rhei.

OBJECTIVE:To evaluate the qualities of Radix Rhei. planted in different areas and study preparing technology and quality standard of Extractum Rhei.METHODS:1. The Rhei collected from different areas was analyzed, according to the quality standard of 2005; 2. The reflux extraction and percolation extraction process of Rhei. were evaluated using orthogonal design, then the preparation of Extractum Rhei. was determined 3. The quality standard of Extractum Rhei. was studied on by layer chromatography, HPLC etc. Chromatographic conditions, system applicability, preparations of reference substance solution, test solution, and methodology were investigated to make the determine methods of Cmodin and Chrysophanol. The Cmodin and Chrysophanol from Extractum Rhei.of 20 batches were determined and analysed.RESULTS:1. Quality analysis from 18 different batches showed:all the quality indexes of 10 batches were qualified according to the pharmacopoeia, rhapontin determination of 5 batches were unqualified, ash content of 2 batches were unqualified, content determination was unqualified of 1 batch. The result showed a great difference among the qualifications of Chinese Medicines. Rhapontin determination faultiness was the most important reason of the failed 8 batches, the Rhei from SiChuan province had the highest percent of pass, followed from GanSu and the lowest from QinHai.2. The results from investigating of the preparation of Extractum Rhei indicated that there was no significant difference in the total content(%) of Cmodin and Chrysophanol. The results of reflux extraction and percolation extraction were A2B1C2 (Etoh%:60%; Dipping time:12h; solvent:8 times) and A3B3C2 (Etoh%:75%; Refluxing time:12h; solvent:8-10 times).3. Methods for analysis of quality, identification, content in Extractum Rhei were determined, and the quality standard of Extractum Rhei. was established.CONCLUSION:The method established in this paper is convenient, practicable and well controlled, and can be used for the inspection of Extractum Rhei.
