Rhei Radix et Rhizoma, which from dry root and rhizome of Rheum tanguticum Maxim.exBalf., Rheum palmatum L. and Rheum officinale Baill., has the efficacy of tapping stagnant,clearing damp-heat and discharging fire, cooling blood and dispelling stasis. Rheum Radixincludes a number of chemical constituents, and has many kinds of pharmacological effect.Medicinal material of Rheum Radix is widely distributed in GanSu, SiChuan, QingHai andYunNan provinces in China, which have large differences in quality. It is essential to establishperfect and comprehensive quality evaluation system for screening fine varieties and ensuringsafe, effective, stable and controllable of medicine.The study was based on the Rheum Radix from different producing areas, and determinatedthe main effective and active component, studyed fingerprint, which to research the qualityevaluation systematically, and to provide reference and basis for further study. This studyincludes the following aspects:(1) To establish UPLC method for determinating eight constituents of Rheum Radix fromdifferent regions as indicator simultaneously, including five free anthraquinones constituents,two Senna glycosides and one stilbene Glycosides. Eight constituents performed good linearrelationship in selected range(r≥0.9996), and the average recoveries were in the range of96.7%~103.2%(n=3), RSD was1.2%~3.2%. Using ultraviolet spectrophotometer(Phenol-sulfuric acid method) to determinate the content of polysaccharide in Rheum Radix. Theresults shown that Rheum tanguticum produced in Qinghai province possessed higher content ofrhubarb polysaccharide relatively.(2) Using UPLC-PDA method to eatablish rhubarb medicinal herbs fingerprint, and obtainfeature maps that could express specificity of the material of rhubarb. Each batch of authenticrhubarb shown higher similarity respectively(S>0.784), and lower similarity with adulterants ofrhubarb (S<0.546), which could be used to rapid identification of rhubarb and its adulterants.(3) The method of1H-NMR was utilized to determinate feature information of rhubarb, andtranslated into data matrix. PCA and DPLS method were utilized to discriminatory analysisbased on the data matrix.1H-NMR fingerprint-pattern recognition could identify samples ofrhubarb in different species effectively.(4) Using UPLC/Q-TOF-MS technology to determinate rhubarb materials, establish mapsto identify eighteen compounds, and find seven compounds that had larger difference, includingsennoside A, sennoside B, rhaponticin. (5) Studying on analysis of structural characters of rhubarb polysaccharide, homogeneouspolysaccharide was obtained that the molecular weight was1.21×104. The constituents ofmonosaccharide in rhubarb were β-D-galactose, α-D-glucose and α-D-xylose, and the mole ratiowas1.1:4.2:1.8. The analysis of HIO4oxidation and methylation shown that main chain ofpolysaccharide was consist of β-D-1→3, β-D-1→4, β-D-1→3,6galactopyranoseandα-D-1→6glucopyranose, and branched chain was consist of α-D-1→terminal xylofuranose.This research utilized the new principle and methods under the guidance of the basic theoryof traditional Chinese medicine, and evaluated the quality of Rhubarb by synthesizing multiplechromatography and chromatography mass spectrometry technology combined with chemicalcomposition pattern analysis method, which could differentiate Rhubarb and its adulterants andevaluate the Rhubarb materials.
