Background and Objective:Renal Clear Cell Cancer(RCCC) is the most common subtype in the Renal Cell Cancer. Biologic behavior of RCCC is of extremely diversity, and there is interindividual variation, It is important to search the prognostic factors of RCCC. The aim of study was to investigate the expression of Dysadherin and E-cadherin, and to analyze their role in RCCC.Methods:40 cases of surgically resected fresh RCCC were collected from 2007-2008 in the department of pathology, The first affiliated hospital of ShanXi Medical University.The control group were cancer-adjacent 3.0 cm of the 40 cases of RCCC, the patients had not been adjunctive therapy before operation. Pathology and clinical data were all. The routine H&E-stained sections were reviewed to verify the morphologic diagnosis. RT-PCR analysis was used to detect the gene expression of E-cadherin and Dysadherin. Western-blot analysis was used to detect the protein expression of E-cadherin and Dysadherin. Statistical analysis was performed using the SPSS 13.0 software.Results:1. Among the 40 cases of RCCC, the expression of Dysadherin mRNA in nuclear gradeâ… -â…¡was lower than that in nuclear gradeâ…¢-â…£(P<0.05), the Dysadherin mRNA expression in the RCCC was higher than in the control group (P<0.05).2. The E-cadherin mRNA expression has no association with nuclear grade in the RCCC(P>0.05). However the E-cadherin mRNA expression in the RCCC was obviously lower than that in the control group (P<0.05).3. The Dysadherin mRNA expression was higher than that E-cadherin mRNA expression in the RCCC (P<0.05).4. The higher Nuclear grade was, the higher the expression of Dysadherin protein and gene was, Dysadherin protein expression of RCCC was higher than that in control group (P<0.05).5. There was only faint E-cadherin protein expression in RCCC. It was higher in the control group.6. The expressions of Dysadherin and E-cadherin have no association with age, sex, and tumor size (P>0.05).Conclusion:1. In RCCC, the higher Nuclear grade was, the higher the expression of Dysadherin protein and gene was, Dysadherin may be become a desired tumor marker of prognostic in RCCC.2. In RCCC, the expression of Dysadherin was higher than that of E-cadherin. There was E-cadherin mRNA expression in RCCC, but There was only faint E-cadherin protein expression. It was likely that Dysadherin expression is one of the mechanisms regulating down-regulation of E-cadherin expression.3. There were the expression of Dysadherin and E-cadherin in the RCCC and control group. In the RCCC, the higher the expression of Dysadherin was, the lower the expression of E-cadherin would be, Dysadherin and E-cadherin may play an important role in the genesis and development of RCCC.
