| Objective1.real-time fluorescent quantitative PCR and immune protein printing was adopted to detect the expression of CK15 and E-cadherin in human esophageal squamous cell carcinoma tissue,the tissue adjacent to carcinoma and normal esophageal tissues.2.To explore the clinical significance of CK15 and E-cadherin in esophageal squamous.Materials and MethodsThe tissues of Esophageal squamous cell carcinoma and its adjacent(The specimens were taken from 2cm and less than the cutting edge of esophageal cancer),and normal esophageal tissues(Specimens taken from 5 cm and more than the margin of esophageal cancer tissue)were collected,60 cases for first and second kind,30 cases for the last kind,from Surgical patients in Taian City Central Hospital of Shandong Province from 2016-01 to 2017-12,and the acquisition of tissue specimens was approved by the relevant department.The standards of Esophageal cancer patients with tissue removed for specimen:(1)Preoperative pathological biopsy and postoperative routine pathology both were diagnosed as esophageal squamous cell carcinoma.(2)Surgical patients are first treated or have seen a doctor but have not been treated.(3)Patients’ previous regular physical examination,no history of other malignancies.All the tissues are stored in a refrigerator at-80°C for sparing.According to the degree of differentiation,there were 22 patients in the well-differentiated group,20 in the moderately-differentiated group,18 in the poorlydifferentiated group,and 0 in the undifferentiated group.Real-time quantitative PCR(RT-PCR)and Western blot were used to detect the expression of CK15 and E-cadherin in all samples.To analyze the expression characteristics of CK15 and E-cadherin in esophageal squamous cell carcinoma,paracancerous tissue,and normal esophageal tissue by using statistical methods(correlation analysis,t-test,etc.).Results1.The relative expression of E-cadherin mRNA in esophageal squamous cell carcinoma tissue was 7.27±2.81,which was significantly lower than that in the pericancerous tissue and normal tissue(7.72±3.04,8.23±3.17).The difference was statistically significant(P<0.05).In the meantime,the relative expression of E-cadherin protein in esophageal cancer tissue was 0.62±0.245,which was significantly lower than that in the tissue and normal tissues of the cancer(0.76±0.297,0.87±0.312).The difference was statistically significant(P<0.05).Compared with normal esophageal tissues,the relative expression levels of E-cadherin-related mRNA in esophageal cancer tissues with different degrees of differentiation were significantly different.2.The relative expression of CK15 total RNA in esophageal cancer tissue was 8.91±3.32,which was significantly higher than that in the pericancerous tissue and normal tissues(8.48±3.18,8.12±2.76).The difference was statistically significant(P<0.05).In the meantime,the relative expression of CK15 protein in esophageal cancer tissue was 1.48±0.282,which was significantly higher than that of paracancerous tissues and normal tissues(1.02±0.213,0.99±0.181).The difference was statistically significant(P<0.05).Compared with normal esophageal tissues,the relative expression levels of CK15-related mRNA in esophageal cancer tissues with different degrees of differentiation were signifycantly different.Conclusions1.The expression of CK15 protein and its mRNA in esophageal squamous cell carcinoma was significantly higher than that in pericancerous tissues and normal esophageal tissues.And the expression in pericancerous tissue was significantly higher than that in normal esophageal tissue.2.The expression of E-cadherin protein and its related gene mRNA in esophageal squamous cell carcinoma was significantly lower than that in pericancerous tissue and normal endometrium.And the expression in the cancerous tissue was significantly lower than that in the normal esophageal tissue.3.The expression of CK15 and E-cadherin is related to histopathological differentiation of esophageal squamous cell carcinoma:the expression of CK15 increased with the degree of differentiation of cancer tissues decreasing,but the expression of E-cadherin was opposite.4.Combined detection of CK15 and E-cadherin expression in esophageal cancer can provide valuable detection indicators for the early diagnosis and prognosis of esophageal squamous cell carcinoma. |
