| Objective:to investigate the relationship of adiponectin (APN) leves to single nucleotide polymorphism at position 276 (SNP+276) and type 2 diabetes-mellitus (T2DM) and carotid artery intima-media thickness (IMT) in Yanbian area in China.Object and method:190 cases were randomly selected as normal group (normal glucose tolerant, NGT), of which 100 cases of the Han, Korean in 90 cases; 217 cases were randomly selected,123 cases in which the Han, Korean 94 cases. according to Carotid IMT217 cases of T2DM were divided into four groups:①arotid atherosclerosis group of Han (IMT≥1.0mm and/or Plaque).②without carotid atherosclerosis of Han(IMT< 1.0mm and without Plaque).ヽarotid atherosclerosis group of Korean(IMT≥1.0mm and/or Plaque).④without carotid atherosclerosis of Korean(IMT< 1.0mm and without Plaque). carotid atherosclerosis group of Han and carotid atherosclerosis group of Korean was divided into three groups, according to the genotype, GG genotype group,GT genotype group, and TT genotype group. serum adiponectin levels were determined by enzyme-linked immunosorbent assay. carotid artery IMT was carotid artery Color Doppler. Using Taqman-MGB probe genotyping, fluorescent sequencing method in gene sequences.AndClinical indicators was recorded. The data were statistically analyzed by SPSS 13.0 software.Results:1. Han and Korean T2DM group serum APN levels of carotid atherosclerosis is lower than on-group, significantly different (P<0.05); carotid atherosclerosis group age, dura-tion is higher than those without carotid atherosclerosis group, significantly different (P< 0.05). 2. Yanbian NGT group and the T2DM group of SNP+276 genotype distribution for the TT (46.8% VS 14.3%), TG (27.9% VS 30.2%), GG (25.3% VS 46.5%); the allele frequency of T/G (60.8% VS 44.7%,39.2% VS 55.3%), two groups of genotypes and allele frequencies were significantly different (P 0.001).3. Yanbian NGT group of Han and Korean SNP+276 genotype distribution for the TT (35.0%V.S35.6%), TG (38.0% V.S 36.7.9%), GG (27.0% V.S27.7%); its allele frequency T/G (54.0% VS 754.4%,46.0% V.S 45.6%), genotype and allele frequency distribution of the two groups no significant difference (P>0.05).4. Yanbian T2DM group of Han and Korean SNP+276 genotype distribution for the TT (17.9% V.S 11.7%), TG (36.6% V.S 41.5%), GG (45.5% V.S 46.8%); its allele frequency T/G (36.3% VS 32.49%,63.8% V.S 67.67%), genotype and allele frequency distribution of the two groups no significant difference, (P>0.05).5. SNP+276 genotype distribution of the TG/TT (48.2% V.S 56.5%), GG (51.8% V.S 43.5%) in of Han in Yanbian area with and without carotid atherosclerosis in T2DM, two groups no significant difference in genotype distribution(P>0.05).6. Yanbian Korean in T2DM with and without carotid atherosclerosis group distribution of gene SNP+276 was TG/TT (50% V.S 58.2%), GG (50.0% V.S 42.0%), two no significant difference in genotype distribution(P>0.05).7. Whether or not carotid atherosclerosis in T2DM group, GG genotype compared with TT genotype, the former BMI, carotid artery IMT and duration were higher than the latter, but the GG genotype in patients with serum adiponectin is lower than the TT genotype (P<0.05).8. Logistic regression indicated:In Yanbian area, the risk factors related to carotid atherosclerosis:age (B= 0.101, OR =1.106, P=0.000), duration (B= 0.065, OR= 1.375, P=0.028), glycosylated hemoglobin (glycosylated hemoglobin, HbA,C) (B= 0.253, OR= 1.290, P= 0.013), APN (B=-0.943, OR =0.390, P=0.000).Conclusion:1.APN levels is correlated to T2DM and carotid artery IMT in Yanbian area.2 SNP+276 T/G are associated with T2DM. in Yanbian area..3. SNP+276 T/G is no difference in the Han and Korean groups of T2DM.4. SNP+276 T/G is no correlate with carotid artey IMT of T2DM.5. APN is a protective factor of T2DM and carotid arteriosclerosis. |
