Objective To explore the association of SNP-11377C>G in promoter region of adiponectin gene with type 2 diabetes mellitus and carotid artery intima-mdia thickness (IMT).Methods The adiponectin gene -11377C>G polymorphism was identified by polymerase chain reaction fragment length polymorphism (PCR--RFLP) in 195 type 2 diabetes (T2DM)patients (include 94 patients with incrassation of carotid artery IMT and 101 patients without incrassation of carotid artery IMT) and 98 nondiabetes normal control subjects. Total cholesterol(TC),serum triglyceride(TG),fasting plasma glucose(FPG),low-density lipoprotein(LDL-C), hight-density lipoprotein(HDL-C)by full automatic biochemical analysor, fasting blood insulin(FINs)and high-sensitivity C-reactive(hs-CRP)were measured by radioimmunoassay,and insulin resistance index(HOMA-IR)was calculated. Serum adiponectin level was assessed by ELISA. Blood pressure (BP), height and body weight were measured, and body mass index(BMI) was calculated.Results 1. The frequencies of adiponectin gene SNP-11377C>G genotype and allele gene were different between T2DM patients without incrassation of carotid artery IMT and T2DM patients with incrassation of carotid artery IMT. There were on differences in genotype and frequencies between subjects with T2DM group and nondiabetes group.2. In T2DM subjects, the values of carotid artery IMT in subjects with SNP-11377CC group was significantly lower than that in subjects with non-SNP-11377CC group, otherwise, serum adiponectin level was higher in SNP-11377CC group than that in non-SNP-11377CC group. 3. Comparison in clinical data between T2DM group and nondiabetes group: Systolic blood pressure (SBP), TG, LDL-C, FPG, and HOMA-IR in T2DM group were significantly higher than those in nondiabetes group, otherwise, serum adiponectin level was lower in T2DM group than that in nondiabetes group.4. Compared with T2DM patients without incrassation of carotid artery IMT, the T2DM patients with incrassation of carotid artery IMT have much higher levels of year, history of diabetes and SBP, but lower adiponectin level.Conclusion In T2DM patients the -11377C>G polymorphism of the adiponectin gene were association with carotid artery IMT, and the values of carotid artery IMT in subjects with SNP-11377CC group was significantly lower than that in subjects with non-SNP-11377CC group. The result suggested that carriers of the G allele gene have a higher risk of type 2 diabetes macrovascular diseases. The -11377C>G polymorphism has no association with type 2 diabetes.
