Screening Of The Natural Anti-tumor Substances In Fruits Of Lonicera Maackii(Rupr.) Maxim

The antitumor activities of different crude extracts from Lonicera maackii (Rupr.) Maxim fruit were studied in the present work. The aim of this work was to find the natural products of antitumor in order to provide the experimental bases to protect and exploit this wide spread resources.The antitumor activities of the different crude extracts from Lonicera maackii (Rupr.) Maxim fruit were measured by MTT in vitro. The results demonstrated that the EtOAC extract of material 1 appeared the highest cytotoxic effects on human cancer cell line SMMC-7721 and HL-60. The IC50 values were about 238.46μg/ml and 299.59μg/ml, respectively. The IC50 of Petroleum ether and water extracts was about 388.46μg/ml and 531.38μg/ml to the cancer cell SMMC-7721. To the cancer cell of HL-60, IC50 of Petroleum ether and water extracts were about 410.68μg/ml and 495.17μg/ml.After further purification of EtOAC extracts of Lonicera maackii (Rupr.) Maxim fruit by column chromatography, nine fractions were collected. Among them, the Fraction2 showed the highest antitumor activities. Fraction3 had the second strong antitumor activities. Fraction2 and Fraction3 inhibitory effect were both much larger than the EtOAC extract, suggesting that further purification and concentration of anti-tumor substances.After further purification to Fraction2 and Fraction3 of Lonicera maackii (Rupr.) Maxim fruit by column chromatography, Obtained from the Fraction2 colorless needle crystal, recorded for the compound 1, but anti-tumor activity of compound 1 was not detected.compound 2 which had strong anti-tumor activity was found in Fraction2 and Fraction3.to the SMMC-7721 cells in vitro, the IC50 of compound was 19.36μg/ml, compound 2 also showed significant activity against HL-60 cells, IC50 is 39.78μg/ml.Compound 1 and compound 2 isβ-sitosterol and oleanolic acid, respectively, by HPLC and NMR spectral analysis, and literature.
