| ObjectiveAim to investigate the immune regulation mechanism and of Chuankezhi (CKZ), a model formula Chinese medicine of warming and recuperating kidney yang Method on allergic asthma treatment.Methods1. Animal Experiment:Mice were sensitized and challenged with ovalbumin (OVA) to establish allergic asthma-like model. To determine the contribution of CKZ on airway allergic inflammation and immune regulation, we examined the BALF cell differentiated count, lung histopathology, cytometric analysis of spleen CD4+CD25+Foxp3+ cells by FCM, and expression of Foxp3, TGF-beta and IL-10 mRNA in the lung tissue by RT-PCR.2. Clinical Experiment:40 mil,moderate or severe asthmatic patients collected from the department of Respiratory Medicine,2nd clinical hospital, University of Guangzhou TCM were randomly divided into control group and Chuankezhi (CKZ) group. The control group received conventional treatment. CKZ group received the intramuscular injection (4ml qd x 7days) besides the routine treatment. The changes of the primary symptoms,PEF values,blood T cell subsets were recorded every other day during the treatment period.ResultsCKZ treatment suppressed murine airway eosinophilia, peripheral vascular congestion and alveolar compensatory expansion, up-regulated spleen CD4+ T, CD4+ CD25+ Treg cells and Foxp3+ CD4+CD25+Treg cells (p<0.05), down-regulated lung tissue IL-10, TGF-P mRNA expression (p<0.01), as compared with asthma group. The clinical improvement was significant including clinical symptom relieving (P<0.05), PEF% increasing (P<0.05) and higher CD4,CD8+T cell percentage in peripheral blood (P<0.05), when compared with control group.Conclusions CKZ treatment suppressed airway eosinophilia, restored Foxp3+ Treg cells in asthma mice model, and relieved clinical symptom, up-regulated PEF% and restored CD4+/CD8+ T cells percentage in asthma patients. Taking together, the study demonstrates an immunotherapy mechanism for allergic asthma by Bu Shen Wen Yang Method raised from traditional Chinese Medicine. |
