Objective:To investigate the expression of ICAM-1 in the retina of rats of hyperhomocysteinaemia.Methods:1.The experiment was lasted for 6 weeks.52 rats divided into groups were used in the experiment. One group (n=26) consisted of animals which were fed a standard diet and given methionine in the drinking water-concentration 1.5g/L. The aim of such a diet was to cause hyperhomocysteinemia. Control group were fed a standard died and clean water.2.We detected the expression of ICAM-1 in the retina of rats by the immunohistochemisty and Western blot.Results:1.Fed rats after 6 weeks, rat tail blood was collected to the determination of Hcy plasma homocysteine. In the control group mean plasma Hcy concentration was 7.01±2.00μmol/L,the model group was 42.91±12.02μmol/L. All the rats in the control group of individual plasma Hcy concentration of less than 15μmol/L, all the rats in model group of individual plasma Hcy levels were higher than 15μmol/L. No rats were removed from the experiment. The T-Ttest, P= 0.000, P<0.05, so the two groups in blood concentrations of homocysteine difference was statistically significant.2.In HE sections, the control group and experimental group no significant change in organizational structure of the retina. The two groups showed no retinal hemorrhage, inflammatory cell infiltration, vascular occlusion and other changes.3.Immunohistochemical examination showed the control group OD value of 0.284 ±0.0383; HHCY group was 0.486±0.0329. There was significant statistical difference between the two groups.4.In the control group, the average of ICAM-1/β-actin ratio was 0.35±0.04. In the model group,the ratio was 0.49±0.03. Two experimental rat retinal tissue In OD values of the Western blot bands of color by the t test, P= 0.000, P<0.05. Therefore, OD values of the two groups was statistically significant difference.Conclusion:1. Methionine concentration of 1.5% of drinking water could induce increased moderately hyperhomocysteinaemia in rats, indicating a successful animal model of manufacturing;2. The ICAM-1 was detected in the retina of rats by immunohistochemistry and Western bolt method;3. Hyperhomocysteinaemia promotes the expression of ICAM-1 in the rats;4. In the experiment all the animals had not been found significant fundus lesions, suggesting that the occurrence of the pathogenic role of hyperhomocysteinemia may also need more time, or combination with other risk factors.
